| Objective:To detect pre-operative and post -operative of serum levels of prostasin in epithelial ovarian cancer and the expression of prostasin in corresponding pathological tissues, and to analyse the relationship between serum and tissues with prostasin..Exploring prostasin as tumor marker alone or with CA-125 inspection to be used early diagnosis and monitoring curative effect, estimating prognosis of the value for ovarian epithelial cancer.Methods:1. The double antibody sandwich Enzyme-linked Immunosorbent Assa (ELISA) technique was applied to detect the serum concentration of prostasin and CA-125.Among the series,Which clinically and pathologically confirmed epithelial ovarian cancer (EOC) patients with a total of 26 cases,40 cases of benign ovarian tumor as a control,30 healthy control women from the medical crowd. The expression levels of prostasin and CA-125 in three groups were compared, respectively. The correlation between preoperative serum concentration of prostasin and different clinic-pathological characters were analyzed. The preoperative and postoperative serum concentration of prostasin in cancer group were analyzed. The correlation between serum concentration of prostasin and tissue expression intensity was analyzed.2. Immunohistochemistry methods were used testing the tissue samples of 20 cases normal ovaries,25 ovarian benign epithelial neoplasm,19 borderline ovarian tumors and 56 cases epithelial ovarian cancer of patients, and to compare positive express situation of prostasin in four kinds organization, And to analyze the masculine expression rate of prostasin and different clinical pathological features of the relationship between them in ovarian tissue.Results:1. The serum concentrations of prostasin in healthy women, epithelial ovarian benign tumors and ovarian cancer preoperative 5.66(4.94 6.37);7.85 (6.50 9.19); 14.07 (10.3,17.60) ug/ml, respectively. The serum level of CA-125 in ovarian cancer,ovarian benign tumor and normal ovarian cases were respectively 11.68 (9.49,13.87),126 (22.69,39.83) and 246.48(167.43,325.54) u/ml. In ovarian cancer group preoperative, the serum levels prostasin and CA-125 were obviously higher than benign tumor group and healthy women, the difference was statistically significant (P<0.01), but there were not statistically significant between healthy women and benign tumor (P>0.05). Ovarian cancer postoperative The serum concentrations of prostasin on 3 days after operation were 7.88 (6.92,8.85) ug/ml, that was obviously lower than preoperative (t=4.673, P=0.000).2. Using the healthy women and benign tumor group as control, the diagnosis value were the best when the prostasin was at the cut-off point which determined from ROC curve. The operativing characteristic-area under the curve(ROC-AUC)of prostasin was 0.867,sensitivity was 84%(22/26), specificity was 81.4%(57/70). When the cut-off point of CA-125 was 35u/ml,its sensitivity and specificity were 76.92 %(20/26) and 78.57%(55/70), respectively. Both sensitivity and specificity were respectively96.73% and 100% when they were combined with both prostasin and CA-125 which were higher than CA-125 alone,(P<0.05).The serum levels of preo perative prostasin was positively related to FIGO staging, grades, types and prostasin protein expression,but was not correlated with CA-125, age, menopausal status (P>0.05).3. There were no expression of prostasin protein in normal ovarian tissues. The expression of prostasin in epithelial ovarian cancer cases was higher than that in benign ovarian epithelial tumor and borderline ovarian tumor (33/56 vs 6/25 and 7/19,P<0.005) and the statistical significance was, dramatic.There were significant differences between benign ovarian epithelial tumor and borderline ovarian tumor. In addition,the positive expression rate of prostasin was associated with organization classification and the clinical stages ect.4. Epithelial ovarian cancer group, the serum level of preoperative prostasin is linear correlation with organization prostasin expressionConclusions:1.The preoperative serum of prostasin was frequently elevated in epithelial ovarian cancer and it dramatically decreased in after surgery. The serum prostasin was higher in advance cancer.prostasin might be a novel marker for clinical diagnosis and prognosis, efficient judgment. Serum prostasin concentration was independent on CA-25 level changes,which hint that have good complementary effect both for ovarian diagnosis. 2. The expression of prostasin in epithelial ovarian cancer cases was higher than that in benign ovarian epithelial tumor and borderline ovarian tumor. The positive expression rate of prostasin was associated with organization classification and the clinical stages,but was not correlated with age, menopausal status. Detection of prostasin might be helpful to estimate manlignant degree of epithelial ovarian cancer. |
