| Hyperplasia of mammary glands is the highest incidence of mammary gland disease. The essence of hyperplasia of mammary gland is mainly because female, progesterone secretion disorder and the breast tissue is stimulated chronically by estrogen, which makes Mammary gland hyperplasia excessive and and instauration is not complete. Western medicine treatment of this disease have certain curative effect. But it's unfavorable long-term drug use and the side effects are big. Traditional Chinese medicine with its better security, stability, small side effects and other special advantage become the current hot of the study of disease of mammary gland hyperplasia. So, the research of mammary gland hyperplasia has important value and significance.This paper firstly established method of Mammary gland hyperplasia pathological model in rats. According to the dating of Estrogen levels and viscera coefficient, the optimal anti mammary gland hyperplasia drugs were selected from three kinds of drugs namely Antlers dish, Medicinal fungi, Plant medicine. Simultaneously, optimized design of medicinal fungi culture conditions were achieved. Eventually, using the test method of uniform design and demonstration test, the optimal dose was selected, which had resistance to disease of mammary gland hyperplasia. Initially, systematic study of chinese medince had done, which had resistance to disease of mammary gland hyperplasia.(1) Adopting intraperitoneal injection of different doses of oestrogen and progesterone acid, animal models was established. By making breast slice of rats and microscopic examination, ideal animal model method of preparation was determined. Rats were given Estradiol Benzoate with administration dosage for 0.8 mg/kg by intraperitoneal injection. Every two days, rats were injected, which included 10 times. Then Instead of injection progestin, rats were given drug 4mg/kg, altogether for injection 4 times. Model success rate is 100%.(2) The preparation of materials drugs(Antlers plate glue, deer's horn deglued and Antlers plate protein) and their role on mammary gland hyperplasia rats had achieved. Antlers plate protein was got by column chromatography and freeze-drying method. Its'molecular weight was determined by SDS-page. By mice pharmacology experiment research, the optimal medicine that had resistance to disease of mammary gland hyperplasia was determined. It's Antlers plate protein.(3) Using the orthogonal experiment design, optimization of medicinal fungus culture conditions was selected. Then fungi polysaccharide were extracted by water extraction and alcohol sink method. The optimal medicine that had resistance to disease of mammary gland hyperplasia was determined. It's Antlers plate protein, Cordyceps militaris mycelium polysaccharides. (4) The preparation of medicine water extraction objects was using thermal circumfluence extraction method. By mice pharmacology experiment research and, dating of Estrogen levels and viscera coefficient, the optimal anti mammary gland hyperplasia drugs were selected. It's Leguminosae, Liquidambar formosana Hance, Curcuma aromatica Salisb.(5) Through the previous test, Five drugs were selectd. Finally, hormone levels, viscera coefficient and histopathological morphology in mammary gland hyperplasia rats was studied. Then the optimal dose was selected. It's fifth group.By uniform design test, formula dose was determined. That might provide certain basis research and development to the drug for mammary gland hyperplasia. |
