Objective: Rukang Prescription is composed of Chinese thorowax root,fritillariae thunbergii,brunellae cum fructu,deerhorn glue,et al. In traditional Chinese medicine, it has the effects to disperse the depressed liver-energy,make one feel happy, promote blood flow and make the blood clot disappear. This research applies the method composed of intramuscular injection of benzogynestry and luteohormone to duplicate the model of rat with hyperplasia of mammary glands. The aim is to verify the curative effect of Rukang Prescription by observing the changes of pathological morphology of rat with hyperplasia of mammary glands,estradiol and progestogen in blood serum,. expression of estrogen receptor of rat with hyperplasia of mammary glands And then discuss the functional mechanism of Rukang Prescription curing mammary gland hyperplasia from the perspective on adjustment of sex hormone and expression of estrogen receptor .Method: 70 healthy Wistar female rats in cleanness grade,75-85 days, 220±20g, after raising by normal diet for a week, were divided into 6 groups randomly:Normal group, model group, Rupixiao control group(RP), TAM control group, low dose of Rukang prescription group (Rukang prescription I group,RI), high dose of Rukang prescription group (Rukang prescriptionⅡgroup,RⅡ). The rats in normal group were treated with the injection of normal saline on the right lower limb, in dose of 0.2ml/d, once a day for 30 days. In other group, those rats were injected estradiol benzoate, in dose of 0.5mg/kg, once a day for 25 days, then injected progesterone, in dose of 5mg/kg, once a days for 5 days. From the thirty-first day, we extracted separately 5 rats from the normal group and the model group by using the random number table, Results: observing pathological sections the by optical microscope, it showed that mammary gland ducts and mamary gland alveoluses were proliferated obviously compaired with normal group. The results indicated that we established the rat MGH model successfully. Atfter modeling successfully, each group adopted group therapy. Each group was drench with Rupixiao suspension, TAM suspension, low dose of Rukang prescription decoction and high low of Rukang prescription decoction, and with distilled water in normal and model group in the dosage of 10ml/kg every day all for 30 days. Fasting 24 hours after last drench, collected blood by decollation, placed them respectively, detected the serum level of estradiol(E2), progestogen(P),. Cut the third pair of breast in aseptic method quickly, slightly .Then wiped one side with filter paper, fixed it in 4 % polyoxymethylene, paraffin emdedded, slice in HE dyeing, observed the mammary histomorphology. The other side is embedded by paraffin and detect the ER by immunity and histological chemistry method. Counting the number of positive cells by electron microscopy and make the analysis of accounts .Results:Part 1:The effect of Rukang Prescription on pathological morphology of rat with hyperplasia of mammary glandsNormal group:Ductal epithelium of breast arrayed regularly. No expansion was in ductal cavity and acinar cavity. The number of lobule of mammary gland and the gland alveolus in lobule of mammary gland were very few.Model group: The breast increased. Hyperplasia of mammary glands is diffuse. Hyperplasia of the Gland alveolus and ductus was in great number. Gland alveolus in lobule of mammary gland was intensive. and inequality of size. Obviously,there was expansion in ductal cavity and acinar cavity. And so much secretion was in intracavity. Compared with model group, in Rukang Prescription I andⅡgroup,the number of the Gland alveolus was few and the secretion was so little. Hyperplasia of the Gland alveolus in Rupixiao control group(RP) and TAM control group was in different degree. However, expansion in ductal cavity and acinar cavity was not obvious The secretion was little .Part 2: The effect of Rukang Prescription on E2 in blood serum of rat with hyperplasia of mammary glandsCompared with normal group, the content of E2 in model group was higher, which had statistical significance (P<0.01); Compared with model group, the contents of E2 in Rukang Prescription I andⅡgroup, Rupixiao control group(RP) and TAM control group were lower, which had statistical significance (P<0.05 ); Compared with Rukang PrescriptionⅡgroup respectively, the contents of E2 in Rupixiao control group(RP) , TAM control group and Rukang Prescription I group were higher , which had statistical significance (P<0.01).Part 3: The effect of Rukang Prescription on P in blood serum of rat with hyperplasia of mammary glandsCompared with normal group, the content of P in model group was lower, which had statistical significance (P<0.01); Compared with model group, the contents of P in Rukang Prescription I andⅡgroup, Rupixiao control group(RP) and TAM control group were higher , which had statistical significance (P<0.05 ); Compared with Rukang PrescriptionⅡgroup respectively, the contents of P in Rupixiao control group(RP) , TAM control group and Rukang Prescription I group were lower, which had statistical significance (P<0.05).Part 4: The effect of Rukang Prescription on expression of ER of rat with hyperplasia of mammary glandsThe cells which dyed in buffy and coarse grain were ER positive cells . The number of ER positive cells in model group was much more than that of normal group.Compared with model group , Rukang PrescriptionⅠgroup and Rukang PrescriptionⅡgroup can repress the number of the positive cells obviously. Furthermore,the number of the positive cells of Rukang PrescriptionⅠgroup and Rukang PrescriptionⅡgroup was much fewer than that of Rupixiao control group(RP) and TAM control group.Compared with model group , the number of the positive cells in Rupixiao control group(RP) and TAM control group was fewer .Compared with normal group, the accumulated points of model group was higher, which had statistical significance (P<0.01); Rukang PrescriptionⅡgroup compared with normal group, had no significance(P>0.1); Compared with Rukang PrescriptionⅡgroup, the accumulated points in Rupixiao control group(RP), TAM control group and Rukang Prescription I group were higher, which had statistical significance (P<0.01) Rukang Prescription I group compared with Rupixiao control group(RP) and TAM control group, had no significance(P>0.1).Conclusion:1 Rukang Prescription can improve the pathological morphology of rat with hyperplasia of mammary glands,which prove that Rukang Prescription can improve and protect the breast in a certain degree.2 Rukang Prescription have certain preventive and therapeutic effects on experimental rat with hyperplasia of mammary glands. Its mechanism may be that it can adjust the ratio of estrogen and progesterone,and expression of ER.
