Objective:To study the effect of Moschus, Borneol and their compatibilities at different ratio on analogous block syndrome model rats, to elucidate partial mechanism of the efficacy and explore the relationship of compatibility at resuscitation.Methods:cerebral edem on rats caused by cold liquid nitrogen, febricity on rat caused by LPS infection were adopted as cold block syndrome model and heat block syndrome model respectively. To study the effect of Moschus, Borneol and their compatibilities at different ratio on temperature of model rats.Heart rate, breathing rate are measured by physicological recorder too. In order to elucidate the mechanism of the efficacy on specific response and nonspecific response to cold block syndrome model rat, Wet Weight Method was adopted to determine ratio of brain water, ELISA was adopted to determine the CRH, ALD, ET, CNP contents, Nitrate reductase assay was used to determine content of NO in the serum. In order to elucidate the mechanism of the efficacy on heat block syndrome model rats, ELISA, ultraviolet absorption method were adopted to determine the IL-1, TNF-a contents in the serum, EB content in the cerebral tissue.Results:(1) results about analogous cold block model:â‘ compared with the control group,the rats'average temperature of the model group, medicine administrated teams decreased significantly but compatibility at 1:1 group (P< 0.05,P< 0.01). Compatibility at 1:1 group's temperature declined slowly compared with the model group's and it significantly declined slowly than solvent group's (P<0.05). The heart rate of model rats is very significantly higher than the control group's(P<0.01). All borneol teams'and compatibility at 1:1,1:2 groups' is significantly lower than the model group's. There are not significance difference of breathing rate between the sham group and model group, medicine administrated groups(P>0.05). The rate of brain water was significantly higher than sham group (P<0.01), It was significantly lower in Borneol group in 66.67mg/kg doses and compatibility group at 1:1,1:3 than those in model group (P<0.05,P<0.01). there weren't significance of the rate of brain water between borneol, Moschus and all compatibility groups (P>0.05).â‘¡CRH, ALD content in serum of model group is significantly lower than the sham group's (P< 0.05) and there weren't significantly difference of CRH content between all medicine administrated group and the sham group (P>0.05). ALD contents of borneol group, compatibility groups at 1:1,1:3 were higher than the model group (P>0.05) and significantly higher than the solvent group's(P<0.05, P<0.01). Compared the sham group, ET and CNP contents of model group were significantly higher (P<0.05) and NO content was significantly lower(P< 0.05) and ET/NO value was tended to increase(P>0.05). Compared with model group, all medicine administrated groups ET content was significantly lower except compatibility at 1:3 group's(P<0.05). The NO contents of borneol groups in 133.34,200.OOmg/kg dosage and compatibility at 1:2,1:3 groups were significantly higher(P<0.05, P<0.01). CNP contents of all medicine administrated groups decrease significantly(P< 0.05, P<0.01) and the compatibility at 1:1,1:2 groups'were significantly lower than the Moschus, borneol in 66.67,133.34mg/kg dosage groups'.(2) results about the analogous heat block model:â‘ The model group rats' average temperature were very significantly higher than the control group's (P<0.01). The average temperature of Moschus group increased more slower than the model group at 1h,1.5h and 2h after medicine was administrated (P<0.05, P<0.01) and other medicine administrated groups'temperature didn't differ significantly from the model group's(P>0.05). heart rate of the model group was significantly higher than the control group's(P<0.05). The Moschus, borneol and compatibility at 1:1 groups' got very significantly lower than the model group's(P<0.01). There weren't any significant difference of the rate of breathing and brain water between the model group's and other medicine administrated groups'(P>0.05).â‘¡Compared with the control group, inflammatory mediators such as TNF-a and IL-1 of the model group increased very significantly(P<0.01). The Moschus, borneol, compatibility at 1:1 could significantly decrease the content of IL-1(P<0.05, P<0.01). But other drug groups didn't have the significant difference (P>0.05). The EB content of the model group got very significantly higher than the control group's(P<0.01) and the EB contents of Moschus, borneol and compatibility at 1:1 groups were very significantly lower than the model group's(P<0.01).Conclusions:â‘ Moschus and compatibility at 1:1 could inhibit decrease of temperature in the analogous cold block model rats in some extent, borneol groups in all dosage, compatibility groups at 1:1,1:2 could decrease heart rate. Borneol in 66.67 dosage and compatibility groups at 1:1,1:3 could reduce the rate of brain water. Meanwhile Moschus, borneol and compatibility at 1:1 groups could also reduce permeability of BBB caused by LPS infection.â‘¡The partial mechanism were rising stress response to RAS and adjusting the balance of contraction and relaxation substance in vein such as ET/NO, CNP in the analogous cold block model rats and inhibiting the release of IL-1 in the analogous heat block model animals.â‘¢Compatibility of Moschus and borneol could protect heart, brain by improving some indicators in cold block model rats and inhibiting the release of inflammatory mediators in the analogous heat block model animals. It was the base of efficacy on resuscitation - indicating block syndrome - going the Heart Sutra. It increased and kept efficacy of Moschus and borneol at different aspects and with " neglecting nature, keeping effect " meaning.The compatibility at 1:1 group had the significant effect on cold block model rats.â‘£The differences between the cold of borneol and heat of Moschus can be expressed by the heart rate, respiratory rate. It related to the pathological state of the body.
