Experimental Research On Correlation Of Regulating Function On Gastrointestine Of The Method Of "Zúsānlǐ (ST36) Treats The Diseases Of Abdomen" And Stomach-Antrum Interstitial Cells Of Cajal (ICC)

Objects:Using functional dyspepsia (FD) rats as research carrier, we intend to investigate the impact on the content and construction of stomach-antrum interstitial cells of Cajal(ICC) after acupuncturing ST 36 (zusdnli) by immunohistochemisty technique, and probe into part of the functional mechanism of treating gastrointestinal diseases by ST 36 (zusdnli) from perspective of the constructional and functional influence on ICC as well as its gastrointestinal motility stimulation.Methods:Fifty-eight male Sprague-Dawley rats were randomly and equally divided into four groups(n=12):Controlled group, Model group, Zusdnli (ST 36) group, Yangling quan (GB 34) group. Manipulate electric-acupuncture on Housdnli (ST 36) and Yanglingquan (GB 34) with thin-dense alternative wave (frequency of15-20HZ, electric volt of 2-4V, electric current of 2mA) for 20mins per day, which kept the local muscle of rats slightly shaken, treat 5 times as a course,2 days interval between courses, four consecutive treatments in total. Semi-solid nutrient paste method was used to obtain the motor functional change of stomach and small intestine; the content of ICC was detected after marking ICC by c-Kit immunohistostaining; and the distribution of connexin 43 and c-kit were observed by SP immunohistostaining and Polink-2-plus with c-Kit antibodies of goats and connexin 43 antibodies of mice.Results:1. The stimulus of clipping tails combining with irregular diet of rats resulted in weight loss and less eating in model group, the interclass difference between the model group and the controlled group possessed statistical significance.2. The gastric emptying rate and intestinal motion rate became slower than that of the controlled group (P<0.05). The gastrointestinal motility of FD rats improved after manipulating electro-acupuncture on Zusdnli(ST 36) and Yanglingqudn (GB 34) (P >0.05). In addition, the curative effect of Zusanli(ST 36) group was better than that of Yanglingqudn (GB 34) group.3. The result of the quantity and ultrastructure of ICC in the gastrointestine of rats:1) The number of ICC of the model group decreased significantly compared to controlled group (P< 0.01), the surface gap of ICC broadened, chimeric connection got loose, the volume of cells became smaller, and the cell membrane was not clear. Junctional constructions between cells were pathologically hurt to be unclear. Junctions among ICC, between ICC and smooth muscle cells or nerve endings reduced significantly. The distribution of Intramural nerves and ICC in the stomach of rats was uneven and their constructions were destroyed and loose. Synaptic junctions were disordered or even could not be connected into network;2) The number of ICC of the Zusdnli(ST 36) group increased compared to the model group (P< 0.05), and decreased compared to the controlled group. Morphological structure of ICC was generally normal, with increased cell surface synapse and visible cell junctions. The pathological damage of gap junction between ICC and smooth muscle cells or nerve endings was less than that of the model group, and their constructions were clear and tight. The pathological damage of distribution of ICC and nerves of cell wall and ecptoma junction was less than that of the model group, Whereas the quantity and ultrastructure of ICC in Yanglingquan (GB 34) group did't improve significantly compared to the model group.Conclusions:1. The stimulus of clipping tails combining with irregular diet resulted in dysfunction of the liver governing the free flow of qi as well as transportation and transformation of the spleen and stomach in model group, then led to functional dyspepsia.2. The gastric emptying rate and intestinal motion rate of FD rats decreased, then electro-acupuncture on Zusanli(ST36) could improve gastrointestinal motility.3. The number of ICC could be increased and ultrastructure of ICC could be recovered after electro-acupuncture on Zusdnli(ST 36), thus their function was recovered, and the motility of stomach and intestine was promoted, which provided more sufficient experiment evidences for the correlation between Zusanli and stomach.
