Objective: To observe 5-FU and ADM combined with X-ray irradiation interfering gene expression of Livin and apoptosis of MCF-7 breast cancer cells.Methods: The IC50 of 5-FU and ADM were detected by MTT in the time of 72 hours. The experiment had four groups: blank control group, X-ray irradiation group, 5-FU + irradiation group and ADM + irradiation group.The Immunohistochemistry and RT-PCR were used to evaluate the Livin protein and mRNA expression,and the apoptosis rate were analysed by flow cytometry.Results: Campare with the blank control group, the Livin protein and mRNA expression of X-ray irradiation group, 5-FU + irradiation group, ADM + irradiation group were reduced, and the apoptosis rate were increased (p<0.05). 5-FU and ADM combined with irradiation inhibited Livin protein and mRNA expression significantly compared with the X-ray irradiation group, and the apoptosis rate of the first two groups were increased. The difference were statistically significant (p <0.05). Conclusion: 5-FU and ADM combined with X-ray irradiation can enhance MCF-7 cell apoptosis, and with a radiosensitizing effect.The mechanism may be related to the down-regulation of Livin gene expression of MCF-7 breast cancer cells.
