| Objective To investigate the effects of Oral Lactobacillus Plantarum supplementation on Liver function, endotoxemia, apoptosis of liver and tight junction of hepatocytes in an experimental model of obstructive jaundice.Method A total of 50 male Wistar rats were randomly divided into five groups of 10 each :â… ,sham-operated;â…¡,bile duct ligation (BDL);â…¢,bile duct ligation +LP (0.2g/5ml/day);â…£, bile internal drainage (IBD);â…¤,bile internal drainage +LP. Hepatocyte apoptosis was quantitated using the transferase-mediated uridine nick end labeling (TUNEL) assay. Portal blood samples for endotoxin measurements were obtained 10 days later. Transmission electron microscopy, RT-PCR and Western blot to observe the ultrastructure and distribution of tight junction associated proteins Claudin-1, Occludin, Claudin-4, ZO-1.Results Compared to the groupâ… ,a significant increase in Hepatic enzyme, endotoxemia, apoptosis (P<0.01). After oral administration of LP the situation alleviated. Bile internal drainage did great help of liver but compared with groupâ…¤the lat has the more effect. The TJ have been evidently destroyed under electron microscope, after oral LP the junction improved. The TJ of groupâ…£is better then the groupâ…¡but the groupâ…¤has the best amendment.
