Klinefelter syndrome is the most prevalent sex chromosome disorder in man. Though Klinefelter syndrome patients were considered as azoospermic patients in the past as well as at present, many literatures have reported that there were sperm in some Klinefelter syndrome patients′semens. Reproductive hormones such as FSH,LH,PRL andT played an important role in spermatogenesis.The ages,mosaic or non-mosaic were important facters affecting the sperm recovery.The semen volume is an important facter in the research of Klinefelter syndrome.There were very few studies on the relations between serum reproductive hormone,semen volume and age.OBJECTIVES: Firstly,we study the relationship between age, serum reproductive hormone, AZF,semen PH and semen volume of non-mosaic Klinefelter Syndrome patients.secondly,the Klinefelter Syndrome is divided into mosaic and non-mosaic.The mosaic Klinefelter Syndrome is the mediacy of non-mosaic and normal people.Then we study the age, sperm density and semen volume in non-mosaic Klinefelter Syndrome. Our study compared the mosaic and non-mosaic Klinefelter Syndrome in semen and reproductive hormone. to better understand the pathogenesis of Klinefelter Syndrome.Last but not least,although some Klinefelter syndrome patients can find sperm in the semen,not all of the sperm is"qualified",so Klinefelter syndrome patients have to check the delelition of AZF.Our study hoped to provide the necessary experimental basis for those Klinefelter syndrome patients who want to have children. It can also offer theoretical foundation for clinical diagnosis and treatment of Klinefelter Syndrome.METHODS:Semen samples and serum samples were obtained from 73 patients of non-mosaic Klinefelter Syndrome and 2 patients of mosaic Klinefelter Syndrome. Their peripheral lymphocytes were cultured and the abnormal karyotypes were analyzed. Reproductive hormones were detected radio immunoassay AZF microdeletion.Different classification methods were compared;serum hormone and genetic analysis was detected;semen parameters were analyzed. Seminal activity of neutralα-Glucosidase and fructose concentration were measured using spectrophotometry.RESULTS:There were 73 cases of 47 XXY karyotypes in 1509 patients with male infertility. There were 2 cases with some sperms in semen. Semen volume in non-mosaic Klinefelter Syndrome patients was significantly lowerer than that in controled groups (P<0.001). There was significantly negative correlation between age and semen volume(r=-0.387,P<0.05). There was significantly negative correlation between semen volume and ratio of LH/T(r=-0.33, P<0.05).CONCLUSION: The incidence rate of Klinefelter Syndrome is 4.84%.There were none of Klinefelter Syndrome patients is AZF microdeletion.Syndrome patients is correlated with age, ratio of LH/T. Low semen volume in non-mosaic Klinefelter Syndrome patients support the notion that patients with an additional X chromosome are androgen insufficient.
