Objective According to WHO reports,there are about 15% of infertile couples of reproductive age,half of them was due to the male infertile.This study aims to analysis the chromosome karyotype and reproductive hormones in 214 patients in our hospital with azoospermia and oligospermia,so as to explore the genetic factors of male infertility.Methods Peripheral blood lymphotytes were isolated from 214 infertile males with azoospermia and oligospermia attending genetic consultancy in Qingdao Women and Children Hospital during Jan 2012 to Oct 2015.The age of the patients were 22 to 54 years old,with an average of 31.2 years.The karyotypes were analysis by stand G-banding,The reproductive hormones including FSH,LH,PRL,E2 and T were detected by electro-chemoluminescence method.Results Among the 214 infertile patients,the stand G-banding showed that the normal chromosome karyotype accounted for 70.09%(150/214),the abnormal sex chromosome karyotype was 18.22%(39/214),the chromosome polymorphism was 9.81%(21/214)and the chromosome balanced translocation carriers was 1.87%(4/214.Sex chromosome abnormalities including 31 cases of typical Klinefelter syndrome,the karyotype is 47,XXY,one case of 47,XXY[46]/46,XX[3]/46,XY[1],three cases of 46,XX sex reversal.one case of a chimeric type of sex chromosome aneuploidy,the karyotype is 45,X[23]/46,XY[27],1 cases of 47,XYY syndrome,one case of Chromosome karyotype for 46,X,+mar[41]/45,X[9],there is one patients with 45,X,22p+.The reproductive hormones levels of FSH and LH in chromosome abnormality group were higher than that in normal chromosome group(P<0.05),while T level showed a trend of drop(P<0.05),E2 had no significant change(P>0.05).Conclusions Chromosome abnormality,especially sex chromosome abnormality could lead to azoospermia and oligospermia.Klinefelter syndrome was the most common chromosome abnormalities.Compared with the normal group,the reproductive hormones FSH,LH and PRL levels in the chromosome abnormality group were significantly evaluated,T level showed a trend of drop.Chromosome aberrations and reproductive hormones play an important role in azoospermia and oligospermia,anaylsis of chromosome karyotype and reproductive hormones could provide guidance for the clinic diagnosis and treatment of infertility.
