0bjective:To evaluate the efficiency of neoadjuvant chemotherapy(NACT) with PF regimen on oral squamous cell carcinoma(OSCC) by examining the protein expression of ki-67,EGFR,KGF,p53 and RARβin the OSCC tissues with immunohistochemistry technique before and after NACT seperately.Furthermore, to confirm its chemotherapy efficiency, provide effective clinical regimen of NACT for OSCC and lay the foundation for further research.Methods:The study group of 20 cases of OSCC were all defined by clinical and pathological diagnoses. The expression of ki-67,EGFR,KGF,p53 and RARβwere evaluated in tumor samples by immunohistochemistry before and after NACT with PF regimen in 20 patients suffered from OSCC.Use image analysis system of HMIAS-2000 to study the positive expression.Datas were analyzed by PEMS3.1 version statistical software, and values of P<0.05 were considered to indicate statistically signifienat.Results:The positive staining was located at nuclei for ki-67,p53and RARβ,cytoplasm or membrane for EGFR, cytoplasm for KGF in OSCC. The expression of ki-67,EGFR and p53 in post-neoadjuvant chemotherapy(35.0%,15.0%,10.0%)were signifycantly lower than those in pro-neoadjuvant chemotherapy(75.0%,45.0%,40.0% ),P<0.05; There was an increase in RARβexpression( 60.0 % vs25.0%, P<0.05) after NACT; The expression of KGF had no significant changes.And there were significant statistical differences of comparison of grade information of ki-67,EGFR and RARβexpression in tumor samples before and after NACT(p<0.05).Conclusion:1. Neoadjuvant chemotherapy can significantly inhibit tumor cell proliferation by reducing the expression of ki-67.2. Neoadjuvant chemotherapy can inhibit the expression of EGFR and prevent its activation pathway, ultimately inhibiting cancer cell proliferation.3. Neoadjuvant chemotherapy can inhibit cancer cell proliferation and promote it apoptosis by reducing the expression of p53.4.The expression of KGF had no significant changes after neoadjuvant chemotherapy, showing that the evaluation of KGF is not sensitive to neoadjuvant chemotherapy.5. Neoadjuvant chemotherapy can induce the expression of RARβsignificantly higher after chemotherapy than that before chemotherapy by inducing cancer cell apoptosis.Overall,as for a small sample study,neoadjuvant chemotherapy with PF regimen is effective to OSCC by inhibitting tumor cell proliferation and inducing it apoptosis.And the expression of ki-67, EGFR, p53 and RARβchanged significantly before and after chemotherapy, more effective response to neoadjuvant chemotherapy.
