| Objective To study the effects and mechanism of fucoidan on rats of type 2 diabetes mellitus.Methods1. Sprague-Dawley rats were randomly assigned into normal control group (n=6) and diabetic model group, which were established by injection small dose streptozocin (STZ) after feeding high glucose and lipids.30 Type 2 diabetic rat models were randomly divided into model control group (n=6), Metformin group (n=6), high dose, middle dose, and low dose fucoidan group (n=6). Intragastric administration to each group for 30 days. General situation were observed during and after treatment.2. To execute the rats after treatment, and detect the level of fasting blood glucose, lipids, insulin, superoxide dismutase, nitric oxide synthase, and free fatty acid. Compared the change of each group. The pathological changes of the rats'liver and pancreas were observed in light microscope.Results1. The model rats established by injection STZ after feeding high glucose and lipids conformed to type 2 diabetes rats. Insulin resistance could occur.2. The rats of normal control group had good mental condition. The weight of the rats in model control group increased during the time of feeding high glucose and lipids. The rats had more sleep and less movement than before. After injection STZ, the symptom of diabetes mellitus occurred, especially the urine. The weight reduced a lot. Compared with the model group, the rats of high dose and middle dose fucoidan group and Metformin group improved the general conditions.3. Fucoidan had significant effect of decreasing FBG and the lipids on diabetic rat, and could increase the activity of SOD, and induce the level of insulin, IL-6, NOS, FFA (P<0.05).Conclusion Fucoidan can improve the general condition and the typical symptom of type 2 diabetes mellitus just like high FBG.. Fucoidan has effect to type 2 diabetes mellitus. |
