Objective:To evaluate the inhibitory action and dose-effect relationship of Qingfei oral liquid on Respiratory Syncytial Virus (RSV), and study the effects of Qingfei oral liquid medicated serum on expression of Transforming growth factor (TGF)-β1, Platelet derived growth factor (PDGF)-BB mRNA genes in Human Embryonic Lung Fibroblasts (HELF) cells infected with Respiratory Syncytial Virus (RSV).Methods:Taking ribavirin as the control group, we used MTT method to observe the inhibition effects of Qingfei oral liquid on RSV among different doses. We attacked in vitro cultured HELF cells with RSV, then we intervened these cells with Qingfei oral liquid medicated serum, Ribavirin medicated serum and blank serum respectively, we determined the expression of Transforming growth factor (TGF)-β1, Platelet derived growth factor (PDGF)-BB mRNA genes with Real-time fluorescence quantitative PCR method.Results:Qingfei oral liquid containing serum on HELF cells maximum non-toxic concentration of 20%, revealed obvious anti-RSV effect, andno significant difference was found when compared with ribavirin. PDGF-BB in the control group increased to 3 times Compared with normal cells, while TGF-β1 decreased to 1/3, Qingfei oral liquid medicated serum group could decrease PDGF-BB to 1/5 of that in RSV infected cells, and TGF-β1 increase to 2 of that in RSV infected cells. Ribavirin medicated serum group and blank serum group had similar effect, they had certain effect on expression of TGF-β1 and PDGF-BB.Conclusion:(1)Infection of RSV can increase the expression level of PDGF-BB mRNA in HELF cells, while decreasing the expression level of TGF-β1 mRNA significantly;(2)Qingfei oral liquid can regulate mRNA expression of cytokines(Such as TGF-β1,PDGF-BB,etc.), and it can attenuate the abnormal changes of RSV infected cells, which may be the mechanism of its antiviral effect. In summation, Qingfei oral liquid can antagonize RSV, and it is an effective and safe compound in treating RSV infections. |
