Backgroud Psoriasis is a kind of chronic,recurrent,difficultly cured and autoimmune-mediated inflammatory disease featured by the main clinical manifestation of erythema,papules with scales on the surface.Traditional Chinese Medicine of every period named the deasease differently such as "ganxuan","songpixuan"and"baikeli",but the syndrome of them is similar.The TCM mechanism of this disease is pathogenic xuefen factors. According to the concensus of TCM experts,we can classify psoriasis by three types named Blood-Heat syndrome, Blood-Xu syndrome and Blood-Stasis syndrome respectively.Recently,trial research showed that psoriasis of Blood-Heat syndrome may be related with T cells mediated autoimmune inflammation,especially Thl cells and Th17 cell. Several researches also revealed that y8T cells may play an important role in pathogenesis of psoriasis.γδT cells can divided into three subsets:effector cells, central memory cells, effector memory cells.They can act like APC cells and secrete cytokines related to Th17 cells.So to investigate the relationship of the functional subsets of y8T cells and psoriasis is vital to reveal the pathogenesis mechanism of psoriasis.Objective In order to study the relationship between the activation and proliferation levels of different subgroup y8T cells and those cells in their different activation stages and the pathogenesis of psoriasis vulgaris with blood-heat syndrome, the project observed the percentage of different y8T cells subgroups and those cells in their different activation stages and analyzed the levels of activation related factors CD38 and HLA-DR and proliferation related factors Ki67 of those cells.Methods Percentage of y8T cell subgroups and those cells in their different activation stages and the activation and proliferation levels of these cells in Peripheral blood (PB) of psoriatic patients with blood-heat syndrome and control groups were dectected by 15 Polychromatic Flow Cytometry.Results (1)In PB of patients with psoriasis vulgaris of blood-heat syndrome,the percentage of CD8-y8T cells expressing Ki67+is(11.60±9.23)%.The percentage of that in the control group is (9.63±10.59)%.Both of them had significant difference(P<0.05).(2)In PB of patients with psoriasis vulgaris of blood-heat syndrome,the percentage of CLA+CD8-y8T cells expressing Ki67+is (42.64±22.13)%.The percentage of that in the control group is (28.78±19.84)%.Both of them had significant difference(P<0.05).(3)In PB of patients with psoriasis vulgaris of blood-heat syndrome,the percentage of effector CD8-y8T cells expressing Ki67+is (17.99±11.46)%.The percentage of that in the control group is (14.23±15.26)%.Both of them had significant difference(P<0.05).(4)In PB of patients with psoriasis vulgaris of blood-heat syndrome,the percentage of CLA+ effector CD8-y8T cells expressing Ki67+is (48.72±26.41)%.The percentage of that in the control group is (35.41±25.07)%.Both of them had significant difference(P<0.05).(5)In PB of patients with psoriasis vulgaris of blood-heat syndrome,the percentage of effector memory CD8-y8T cells expressing Ki67+is (13.75±13.03)%.The percentage of that in the control group is (7.76±6.78)%.Both of them had significant difference(P<0.05).(6)In PB of patients with psoriasis vulgaris of blood-heat syndrome,the percentage of effector memory CD8-y8T cells is (46.29±27.59)%.The percentage of that in the control group is (31.43±26.69)%.Both of them had significant difference(P<0.05).(7)In PB of patients with psoriasis vulgaris of blood-heat syndrome,the percentage of CLA+ effector memory CD8-y8T cells is (15.84±18.90)%.The percentage of that in the control group is (15.54±29.95)%.Both of them had significant difference(P<0.05).(8)In PB of patients with psoriasis vulgaris of blood-heat syndrome,the percentage of central memory CD8-y8T cells is (27.49±13.07)%.The percentage of that in the control group is (41.23±19.03)%.Both of them had significant difference(P<0.05).(9)In PB of patients with psoriasis vulgaris of blood-heat syndrome,the percentage of effector memory CD8+y8T cells is (29.26±18.58)%.The percentage of that in the control group is (18.44±18.32)%.Both of them had significant difference(P<0.05).(10)In PB of patients with psoriasis vulgaris of blood-heat syndrome,the percentage of CLA+ effector memory CD8+y8T cells expressing CD38+HLA-DR+is (20.26±25.65)%.The percentage of that in the control group is (6.75±9.15)%.Both of them had significant difference(P<0.05).(11)In PB of patients with psoriasis vulgaris of blood-heat syndrome,the percentage of CLA+ effector CD8-γδ17 cells expressing Ki67+is (34.34±25.40)%.The percentage of that in the control group is (9.64±43.11)%.Both of them had significant difference(P<0.05).(12)In PB of patients with psoriasis vulgaris of blood-heat syndrome,the percentage of effector memory CD8+78T cells expressing CD38+HLA-DR-is (6.26±5.48)%.The percentage of that in the control group is (9.92±7.75)%.Both of them had significant difference(P<0.05).Conclusion The pathogenesis of psoriasis vulgaris with Blood-Heat syndrome may be related with CLA+γδT CD8-y8T cells.This subset of y8T cells may be activated by foreign antigens and stimulated to proliferation.Then antigens are presented to Th17 cells and stimulate the activation of Thl7 cells for the expression of IL-17. CLA+CD8-y8T cells can secrete Thl7 cytokines,too. |
