Objective:The aim of this study is to observer the anti-oxidant avtivities of To ngmai Jiangzhi Pill on diabetic macrovascular disease animal models with aorti c endothelial oxidative stress state.Method:80 male Wistar rat at 5 month old were divided into 4 groups(control group,model group, Tongmai Jiangzhi pill group and Atorvastatin group, respectively)Model group, Atorvastatin group and Tongmai Jiangzhi pill group received nitric oxide synthase(e NOS) inhibitor(L-NAME) 0.lmg.ml-1 diluted in drinking water and high fat diet for 35 days to establish the type II diabetes model. Control group were fed with normal diet. Control group, model group were dosed with 5ml·kg-1·d1 normal saline at the same time, the Atorvastatin group received atorvastatin calcium statin1.6mg·kg-1·d-1 orally and traditional Chinese medicine group were treated with Tongmai Jiangzhi pill 5ml·kg-1·d1 diluted in normal saline. During the experiment, the general state of animals, the amount of drinking water and food intake were observed.The body weight and blood glucose were measured once a week. After 35 days in the breeding, of rats in each group were finished testing blood glucose, and serum was prepared for anesthesia in 2ml/kg intraperitoneal injection of 8% Pentobarbital Sodium, tissue samples of serum total SOD activity detection, detection of serum glutathione peroxidase activity and serum total antioxidant capacity, serum malondialdehyde detection, p47 phox gene detection.Results:1 Compared with the control group, the levels of blood glucose were higher in model group(P<0.01). The model group significantly incrased the levels of SOD,GSH-Px and T-AOC in serum as compared to the control group(P<0.01).2 The levels of serum SOD, GSH-Px, T-AOC and blood glucose were decreased in Tongmai Jiangzhi pill group and Atorvastatin group compared with the model group(P<0.01, respectively). Moreover, the Tongmai Jiangzhi pill group and Atorvastatin group decreased the activities of serum MDA and down regulated the p47 phox expression in blood viscule(P<0.01, respectively).Conclusion:1 The administration of itric oxide synthase(e NOS) inhibitor(L-NAME) 0.lmg.ml-1diluted in drinking water and high fat diet for 35 days could establish the type II diabetes model in rat.2 The Tongmai Jiangzhi Pill could improve the macrovascular disease in diabete patients, the possible machnism may due to the increase of anti-oxidant activities.
