Our libratory using the BIBAC library of ginseng genome, Select the tomato varietiesFenguan888as receptor material, The use of Agrobacterium-mediated leaf disc method forgenetic transformation of tomato. In order to obtain transformed plants, and provide technicalsupport for the identification of ginseng gene function. Ginseng (Panax ginseng C.A.Meyer)belongs to Araliaceae Panax plant. It is a perennial herbaceous plants. The ginsenoside is one ofPanax ginseng’s main effective compontens. Due to less ginseng genetic research, Study of thegenomics is poor, the condition limits depth-study in modern genetics and breeding of ginseng.Tomato (Lycopersicon escaleutum. Mill) belongs to Solanaceae Lycopersicon plant. It is annualor perennial. The main active substance is lycopene. It have the prevention of vascular aging,cancer prevention, anti-aging effects. Tomatoes are dicots, capable of self-pollination,chromosome maps more clearly, and more offspring, reproduction coefficient, easy to operategenetic transformation, easier cultivation etc. So it is one of the most studied model plantbiology. The results of this study are as follows:1.Agrobacterium containing large fragment DNA(D8) COR308logarithmic phase for10to13hours, choose11-12hours of agrobacterium expriment;Agrobacterium containing largefragment DNA(D21) COR308logarithmic phase for10to14hours, choose12-13hours ofagrobacterium experiment.2.Inoculate mediun:MS(1%sucrose);Pre-culturen medium:MS(3%sucrose)+IAA0.2mg/L+6-BA1~2mg/L;Pre-culturen medium:MS(3%sucrose)+IAA0.2mg/L+6-BA1~2mg/L+AS2.0mg/L;Inducing rooting(3%sucrose)+NAA0.1mg/L.3.Transforming conditions is determined as the resuspension liquid of OD550=3.0Agro-bacterium, resuspension liquid component MS(3%sucrose、IAA0.2mg/L、6-BA1~2mg/L).Pre-culture time for tomato genetic transformation is2d, infection time is8min, total trainingdays for4d.4. PCR detection, part of the transformation of plants were positive. ginseng large fragmentof DNA integration into the Tomato genome.5.Through analysis the transformed plants and untransformed tomato plants morphological,we find that transformation plants(D8) in stem, seed and leaves form has changed, veins turninto purple,sterm become smooth and the fluffy become less, the seed of fruit become heavy and small. that transformation plants(D21) in stem, flower and sepals form has changed, leavesand veins turn into purple, the number flower and sepals has changed.
