In our study,the ginseng Ppase were systematically analyzed and identificated by bioinformatics base on the 248,993 unigenes database.The PgPpase involved in ginsenoside biosynthesis were predicted.The expression of PgPpase03-1 gene and the key enzyme gene of ginseng saponin were analyzed which proved accuracy of bioinformatics predictions.Genetic transformation of PgPpase03-1 gene was conducted.Lycopersicon esculentum was used as material:1.In the present study,One hundred and tweenty six PgPpase were screened by bioinformatics method using the constructed 248,993 unigenes database.25 PgPpase have complete ORF and conserved domains.14 PgPpase were predicted to have 10 motifs with MEME.2.Evolutionary analysis show that PgPpase,under different gene ID,are congregated together.Based on phylogeny tree with PgPpase from other phyla,PgPpase have been divided into three distinct clades.PgPpase01-1,PgPpase07-1,PgPpase06-1,PgPpase06-5gene and Ppase of Ostreoccus Tauri,Ostreoccus Ostreoccus Tauri and Chlamydomonas reinhardtii are congregated toghter.It can Indicate that these PgPpcase appeared earlier in ginseng.3.The expression patterns of PgPpase were different in different tissues,different growth environments and development stages.But the PgPpase02-3,PgPpase02-6,PgPpase03-1,PgPpase06-1 and PgPpase06-5 genes are present in all tissue,different growth period and all growth environment.The expression of PgPpase03-1 gene is the highest which much higher than other genes.4.Prediction of genes that may have an effect on the synthesis of ginsenoside,it is concluded that PgPpaste03-1 gene has the most further research value.5.The result of different hormone induced ginseng hair-root showed that the ginenoside have significant difference compared with control,while PgPpase03-1 which expression patterns were similar with key enzyme genes in MVA pathway.The ginseng saponin has improved which proved accuracy of bioinformatics predictions.6.Genetic transformation of PgPpase03-1 gene was conducted.Lycopersicon esculentum was used as material.The conversion rate of the hypocotyl was 5.7 %-8.2 %.The conversion rate of cotyledons was 11.0 %-14.1 %.T0 seeds of 34 positive tomato plants were obtained.
