Ginseng,Panax ginseng C.A.Meyer,is a perennial herb belonging to Araliaceae,which is a commonly used traditional chinese medicine,and was praised by many TCM Classics.Ginseng contains a variety of ingredients,including saponins,polysaccharides,fatty acids,volatile oil et al.Ginsenoside as a major bioactive ingredient in ginseng has been proved to play an important role in a number of aspects in human medicine and health.Despite the high medicinal value of ginseng and the biological activities of ginsenosides,the cultivation cost of ginseng is expensive because of the long growth cycle and serious diseases along with the wild resources exhausted.Therefore,it is urgent to reduce the cultivation cost of ginseng saponins and protect ginseng germplasm resources under the condition of meeting the market demand.One of the most important solutions is to use biotechnology.Homeodomain leucine zipper(HD-Zip)protein is a transcription factor specific to plants,involved in specific biological processes such as photosynthesis and plant morphogenesis,which plays a crucial role in plant growth and development.This family has been studied thoroughly in Arabidopsis,and has not been reported in ginseng.In this study,the HD-Zip gene family of Jilin ginseng was analyzed by bioinformatics method,and the related gene cloning and genetic transformation were carried out,The results are as follows:1.The result of systematic analysis of Jilin ginseng HD-Zip gene family was as follows:(1)Through the method of local database comparison,96 HD-Zip Unigenes were screened from Jilin ginseng 248,993 Unigenes,constitute the Jilin ginseng HD-Zip gene family,and these genes were divided into four types of I,II,III and IV by the conservative structure;(2)The analysis of evolutionary showed that the origin of this family was relatively early and the evolution was conservative;(3)Blast2GO functional annotation showed that these genes were annotated to 11 functional categories on level 2,mainly center on the function of binding,transcriptional activation and cell classification.And it was significantly enriched in all GO annotation functions relative to the controls;(4)The analysis of expression patterns showed that the gene expression of this family was spatiotemporal specific;(5)Network analysis showed that the family genes exist in the form of functional groups,and the genes interact with each other in a cluster way.2.HD-Zip gene PgHB14-2 which related to ginsenoside biosynthesis was cloned and the recombinant plant expression vector pBI121-PgHB14-2 was constructed.3.The tomato inbred P01 D was choosed as a receptor for genetic transformation because of its good regeneration ability,and the gene of PgHB14-2 was transfered into it by Agrobacterium mediated method,finally obtained positive plants.The structure,evolution,functional differentiation and expression patterns of the HD-Zip gene family of Jilin ginseng obtained in this study provide a reference for the excavation and identification of the functional genes of Panax ginseng.
