Objective:To research the influence of astragalus polysaccharids(APS) on CO concentration in blood and testicular micro-structure for varicocele (VC) rats;To indicate the spermatogenesis and sperm quality indirectly.;To explore the predictive index for spermatogenic tissue injury earlier in clinic for VC patients.Methods:80 adolescent male SD rats were randomly assigned to sham-operation group (10 rats) and model group(70 rats). The 70 rats in model group were randomly assigned to astragalus polysaccharide group (40 rats), saline group(20 rats) and control group (10 rats). The 40 rats in astragalus polysaccharide group were assgined to high dose 4-week group, high dose 8-week group, low dose 4-week group and low-dose 8-week group randomly based on the difference of herbal time and administration of herbal concentration when all the modle rats were ready and there were 10 rats in each group. Kill the rats in the 4-week groups and collected left testis and blood from spermatic vein 3ml then centrifuged and got the supernatant 4 weeks later. Detected the absorbance of CO in supernatant with ELISA method and observed the morphological changes of inside diameter of contorted seminiferous tubules, altitude of seminiferous epithelium, spermatogenic cells in different stages and limiting membrane with lumen in microscope.Kill the rest rats in the rest groups and got specimen with the same method like doing in the 4-week groups 8 weeks later.Results:There was statistic difference in the injury of inside diameter of contorted seminiferous tubules, altitude of seminiferous epithelium, spermatogenic cells in different stages and limiting membrane in every 2 groups (P<0.05) except the high-dosage-8-week group and sham-operation group(P>0.05). About the injury of the 3 indexs the rats in the APS groups and low-dosage groups were superior than control group and saline group(P<0.01). The absorbance of CO in spermatic vein blood was higher than before for the modle rats.About the absorbance of CO the rats in APS groups,high-dosage groups,8-week groups were inferior than that in control group and saline group,low dosage group and 4-week groups pespectively. There was statistic difference in the absorbance of CO in every 2 groups(P<0.01).Conclusion:There were protective effect for APS on testis damage caused by varicocele and related to the herb concentration and time. The spermatogenic dysfuction may correlated with the concentration of CO. The concentration of CO could be the susceptibility index for spermatogenic tissue injury earlier in clinic for VC patients. Astragalus polysaccharide can reduce the concentration of CO in blood of rats caused by VC.
