Objective: To observe the drug-containing serum of RAS that is one ministerial drug of recuperating-heart decoction effect the HUVEC oxidative damage model by H2O2induced, to investigate the potential mechanism of to action about it ,to research mechanism of to action about recuperating-heart decoction for prevented and cured UA(unstable angina), to provide objective basis for clinical application.Method: To apply prepared method of drug-containing serum, the 30Wistar rabbits are divided into two groups randomly that blank group and RAS group. the rabbits are gived SLR decoction in the RAS group, the rabbits are gived isodose physiological saline in the blank group. intragastric administration is 2 times a day and continued a week. blood collection is from heart of rabbits at 1 hour latter of the last administer. Then to separate blood serum and to prepare 2%,4%,6% drug-containing serum culture fluid. To apply cell culture method, the refrigerant saved HUVEC is revived, cultivated and passed generation. The cells were digested, collected and counted until the cell condition stabilized. The cell suspension were transfered in culture plank with 96 hole, there are every2×104cells in every hole. It was divided five groups. They are H2O2 model group, blank blood serum group, 2% drug-containing serum of RAS group, 4% drug-containing serum of RAS group, 6% drug-containing serum of RAS group. There are 6 annexal hole in every group. It is interfered in drug-containing serum with different concentration for 24hours in every drug-containing serum of RAS group. It is cultivated with common culture solution for 24hour in H2O2 model group and blank blood serum group. then HUVEC is interfered in 100μmol/L H2O2 for 2 hours(except blank blood serum group).to copy the Atherosclerosis cells model. morphological variation is observed by inversion flourescence digital imaging microscope. The cells viability is measured by MTT assay. Result :1,The morphology result of common cell indicated that HUVEC growth well with monoptychial adherence. It appeares globular and ellipse at beginning. It appears fusiform following. It appeares typical slabstone or cobble-stone. The passage cell expands quicker and adheres paries earlier than primary cell.2,The morphology result of every handling group indicated that in every handling group ,the cells damage obviously with volume shrink and interspace widen appear cell debris. it display disorderly with cytomorphosis and shrinkage. while in groups of 2%,4%,6% drug-containing serum of RAS ,condition of the cell is improved comparing with the H2O2 model group. The cell ablates lessly and cytomorphosis and shrinkage relieves in different degree.3,The result of MTT indicated that The OD value of HUVEC increased obviously in every drug-containing serum of RAS group, comparing with blank blood serum group and H2O2 model group, it was considered statistically significant (p<0.01) every drug-containing serum of RAS group can increase the growth rate of HUVEC by H2O2 damage, comparing with the H2O2 model group, it was considered statistically significant (p<0.01).Conclusion: There are antioxidation in the drug-containing serum of RAS. It can protect the integrality about structure and function of endothelial cell, it can reverse and improved functional damage of endothelial cell, it can protect endothelial cell from oxidative damage. this is maybe the one of mechanism of action for prevented and cured UA.
