OBJECTIVE:To explore the polymorphic 1166A/C of angiotensinⅡtypeⅠreceptor (AT1R) and the polymorphism -344T/C of aldosterone synthase gene (CYP11B2) for association with essential hypertension.METHODS:Association analysis was employed and the two single nucleotide polymorphisms were genotyped by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis of genomic DNA from 100 patients with essential hypertension (HT) and 100 normotensive (NT) Han subjects in Hunan.RESULTS:AT1R 1166A/C and CYP11B2-344T/C genotypic freq-ency did not deviate from Hardy-Weinberg equilibrium. Essential hypert-ension AT1R1166A/C AC type and C allele frequency is higher than no-rmotensive group (chi (2)=4.879; 4.796, P<0.05); essential hyperten-sion blood pressure, triglyceride and cholesterol levels were normotensive group (P<0.05), while the low-density lipoprotein and high density lipop-rotein was no significant difference (P>0.05); AT1R1166A/C AC geno-type of blood pressure, triglyceride, cholesterol, LDL and other clinical parameters were significantly higher than AA genotype, while the AC ge-notype was significantly lower than the high-density lipoprotein AA gen-otype. There was no significantly difference of the distributions of three genotypes (TT, TC, and CC), T and C alleles between the hypertensive and the normotensive groups (chi (2)=0.091,0.022, P> 0.05). The fre-quency of T allele was higher in both groups (HT:63.0% vs 37.0%, P<0.05; NT:64.0% vs 36.0%, P<0.05). Association analysis, compared to AA-TT genotype combined, AC-TC genotype combination had an odds ratio for elevated BP of 2.25CONCLUSION:AT1R 1166A/C was significantly associated with essential hypertension,CYP11B2-344T/C polymorphism is not associated with essential hypertension. With AT1R1166A/C AC-type gene and the AC-TC genotype, People increase the odds of an developing higher BP, Which may play an important role in the development of essential hypertension.
