| Objective: To detect the Notch receptors and ligands gene and protein expression in breast cancers and hyperplasia lesions, and their relationships with clinical indicators of breast cancers and hyperplasia lesions were analyzed.Methods: Notch1 and JAG1 mRNA of 60 breast IDC and 25 normal breast tissues were amplified by one-step RT-PCR method,and the expression of Notch1,Notch3,JAG1 and DLL4 protein was detected by immunohistochemical Streptavidin-Biotin Complex (SP) stain in 90 breast IDC,90 breast hyperplasia lesions and 20 normal gland samples for which formalin-fixed, paraffin-embedded (FFPE). All data were analyzed using the SPSS 13.0 software package.Results: 1. RT-PCR showed: (1)The expression rate and standardized coefficient of Notch1 in breast cancers were significantly higher than those of normal breast tissues. The expression rate of JAG1 in breast cancers was 10%,while JAG1 was not detected in normal breast tissues. (2)The standardized coefficient of Notch1 in cases with axillary node metastasis was significantly higher than that in cases without axillary node matastasis. (3)The standardized coefficient of Notch1 at stage I was significantly lower than that at stage II, and stage II was significantly higher than stage III. There was no statistically significant difference between stage I and stage III. (4)The standardized coefficient of Notch1 at grade I was significantly lower than that at grade II, and grade II was significantly lower than grade III. (5) As a result of fewer breast cancers of the group, we can't compare with Notch1 and JAG1 mRNA expression and clinical indicators. 2. IHC showed: (1)The expression of Notch protein in breast cancer and hyperplasia lesions in both different frequencies. (2)The expression rate of Notch1 was higher than Notch3 in breast cancer, and the expression rate of DLL4 was higher than JAG1 in breast cancer. (3)There was higher statistically difference when Notch3 expression rate of those examples expression Notch1 at the different levels and there was also higher statistically difference when Notch1 protein of the examples coexpressing Notch3 and Notch1 protein was compared with that of the examples expressing Notch1 but not Notch3 protein. (4)The expression of Notch protein in breast cancers have high statistically significant with axillary node metastasis, different TNM stages and pathological grades. (5)There was no statistical difference between the expression rates of ER,PR,C-erbB-2 and Notch protein.Conclusions (1)Abnormal expressing Notch singnal implicated in the formation of breast cancer. Notch functions differently in different progression in breast cancer. (2)Notch signaling protein expression in breast cancer and hyperplasia lesions have different levels. (3)There seems to be a expression relationship between Notch1 and Notch3;Notch1 and DLL4 may play a important role in breast cancer. |
