| Objective:With HC-A comparison of renal preservation solutions to study the self-AMU hospital of multiple organ preservation solution during cold storage of rabbit kidney biochemical indicators of energy.Methods:Rabbit kidney in vitro model of simple cold storage, with the HC-A comparison of renal preservation solution, the control group with 0-4℃HC-A preservation solution for kidney perfusion and preservation were simple, the experimental group made use of 0-4℃AMU preservation solution for kidney perfusion and preservation were simple,24,48,72 h, respectively, after detection of the following indicators.1, renal cortical mitochondrial respiratory control rate (RCR) was detected to the Franco-Prussian detection system through a very testing.2, renal cortical Na+-K+-ATPase activity was examined, to detect biochemical assay.3, renal cortex mitochondria in the calcium content, application of biological and chemical methyl thymol blue method for testing.Results:(1)Respiratory control ratio (RCR)changes:Respiratory control ratio changes were decreased both in AMU group and HCA group. The contents of Histological changes no significant difference between group AMU and group HCA.(P>0.05); at 48h,72h The contents of Histological changes in group AMU was significantly higher than the group HCA(P<0.05). (2) The activity of Na+-K+ATPase in renal cortex were decreased both in AMU group and HC-A group.At each time point,there was no significant difference in the activity of the activity of Na+-K+ATPase between AMU group and HC-A group at 24h and 48h(P>0.05):The activity of Na+-K+ATPase in AMU group was higher than that of HC-A group and there was significant difference at 72h(P<0.05). (3)The contents of mitochondrial calcium:The contents ofmitochondrial calcium were creased both in AMU group and HC-A group.The contents of mitochondrial calcium no significant difference between group AMU and group HCA.(P>0.05);At 48h 72h,The contents of mitochondrial calcium in group AMU was significantly lower than the group HC-A(P<0.05)Conclusion:1, AMU solution for the conservation of the integrity of rabbit renal cortex mitochondria and oxidative phosphorylation coupling liquid level is superior to HCA.2, AMU solution on preservation of rabbit renal cortical Na+-K+-ATPase activity and kidney can be better than for HC-A solution.3, AMU solution on preservation of rabbit renal cortex mitochondria increased activity of calcium to prevent calcium overload better than HCA solution.4, AMU rabbit kidney during cold preservation solution on cellular energy metabolism in renal cortex of the protection is better than HC-A solution is good, with clinical development applications. |
