It becomes the female malignancy with highest incidence. 20(R)-ginsenoside Rg3 (SPG-Rg3) is tetracyclic triterpenoid saponin, present in a natural medicine Ginseng. The anti-tumor effect of the medicine is been pay widely attention, but the mechanism of anti-tumor in breast cancer is to be studied.Objective:To detect the growth inhibiting induced by 20(R)-ginsenoside Rg3 (SPG-Rg3) in human breast cancer MCF-7 cells; to detect the apoptosis induced by SPG-Rg3 in MCF-7 cells, and the effect on cell cycle of MCF-7 cells; to detect the expression of Fas and FasL in MCF-7 cells, and to analyse the correlation between apoptosis and the expression of them.Method:Human breast cancer MCF-7 cells were divided into blank control group, experimental control group and groups with multiple concentrations of SPG-Rg3. MTT was adopted to observe the restraining growth effect intervented by SPG-Rg3 in 24h,48h and 72h, and the IC50 was calculated in 48h to determine the effective concentration; the cycle of MCF-7 cells was detected by flow cytometry after the role of SPG-Rg3; Annexin V- EGFP / PI double staining was adopted to detect apoptosis of MCF-7 cells induced by SPG-Rg3; immunocytochemical was used to detect the relationship between apoptosis of MCF-7 cell and the expression of Fas and FasL.Results:1,MTT result demonstrated that the growth of MCF-7 breast cancer cells induced by SPG-Rg3 was significantly inhibited in 24h, 48h and 72h, and the inhibition rate increased with the increase of its concentration, inhibitory concentration 50 percent (IC50 ) in 48h was calculated to 244.54μg/ml. 2, Flow cytometry results showed that 300μg/ml 20 ( R) - the role of ginsenoside Rg3 in MCF-7 cells 48 hours after S phase cells increased significantly (P <0.01), G2-M phase cells decreased significantly (P <0.05), MCF-7 cell apoptosis was significantly increased (P<0.01). 3, Immunocytochemistry showed that: in experimental groups, expression of Fas and FasL cytochrome C was significantly higher (P <0.01).Conclusion:SPG-Rg3 can induce apoptosis in MCF-7 breast cancer, and increase the expression of Fas and FasL protein.
