Objeceive:Flos Robiniae Pseudoacaciae is the dried flowers of Robinia psendo-acacia L,Through isolating and identifying the chemical constituents of the part of butanol of Flos Robiniae Pseudoacaciae,and the obtained compounds were screened for pharmacological activity, Find the material basis of anti-tumor activity, For the Flos Robiniae Pseudoacaciae development and utilization of the resources spent to provide the scientific basis.Materia and method:1. Materia: Flos Robiniae Pseudoacaciae,the dried flower and bud of pod-bearing plant of Robinia psendo-acacia L.2. Method:2.1 Using the methods of silica gel column chromatography, gel column chromatography and recrystallization , to separate and purify chemical compounds from the part of butanol of Flos Robiniae Pseudoacaciae. Using the physicochemical property,NMR and MS, to identify their structures.2.2 The pharmacological experiment: MTT reduction by 5 kinds of screening compounds Robinin structure etc on human hepatocellular carcinoma SMMC-7721 cells, human gastric cancer MGC-8 cells and human colon cancer HT-29 cell apoptosisResult:1. To obtained 14 chemical constitutes from the part of butanol of Flos Robiniae Pseudoacaciae.By spectral analysis, their structures were respectively N-2-(1, 3, 4 - trihydroxy - octadecyl)-α-hydroxy -3 '- ethylene - tetracosane amide, kaempferol, soya sapogenol B, D-3-O-methylinosit, Kaempferol-3-O-β-D-galactopyrano-side, kaempferol-3-O-β-D-galactopyranose-7-O-α-L-rhamnopyran-oside,kaempferol-7-O-α-L-rhamnopyranosid, pentacosanoic acid, Robinin structure, 4′-methoxy-5,7-dihydroxylflavone-7-O-β-D- glucose residue-(6â†'1)- rhamnoside(linarin), Soy Saponineâ…¢, daucosterol,β-sitosterol, Kaempferol.2. Isolated from the Flos Robiniae Pseudoacaciae isolated kaempferol-7-O-α-L-rhamnopyranosid, Robinin structure, Soy Saponineâ…¢, linarin, N-2-(1, 3, 4 - trihydroxy - octadecyl)-α-hydroxy -3 '- ethylene - tetracosanoic five compounds amide SMMC-7721 human hepatoma cells, human gastric cancer MGC-8 cells and human colon cancer HT-29 cells have different degrees of inhibition, in which Soy Saponineâ…¢on Inhibition of three cancer cells, the rate of 81.5% (human liver cancer SMMC-7721), 84.9% (human gastric cancer MGC-8) and 84.8% (human colon carcinoma HT-29), followed by the locust glucoside, rhamnose Locust glycosides, N-2-(1, 3, 4 - trihydroxy - octadecyl)-α-hydroxy -3 '- ethylene - tetracosane amide and linarin.Conclusion:1. 14 chemical constitutes from the part of butanol of Flos Robiniae Pseudoacaciae are separated and identified, including that N-2-(1, 3, 4 - trihydroxy - octadecyl)-α-hydroxy -3 '- ethylene– tetracosane amide is a new compound , Soy Saponineâ…¢, 4′-methoxy-5,7-dihydroxylflavone-7-O-β-D- glucose residue-(6â†'1)- rhamnoside(linarin) ,soya sapogenol B, pentacosanoic acid are obtained for the first time from Robinia pseudoacacia L.2. MTT method using isolated from Flos Robiniae Pseudoacaciae of the five compounds were screened in vitro antitumor activity, The results of Soy Saponineâ…¢and Robinin structure on SMMC-7721 human hepatocellular carcinoma cells, human gastric cancer MGC-8 cells and human colon cancer HT-29 cells were inhibited well, kaempferol-7-O-α-L-rhamnopyranosid on human hepatocellular carcinoma SMMC-7721 cells and human colon cancer HT-29 cells more sensitive, N-2-(1, 3, 4 - trihydroxy - octadecyl)-α-hydroxy -3 '- ethylene– tetracosane amide on human gastric cancer MGC-8 cells sensitive, The above linarin 3 weaker sensitivity of cancer cells.
