OBJECTIVES:To study on abnormal expression of p53 protein,PCNA and E—cadherin in tumor tissues of patients with papillary thyroid carcinoma (PTC) may have some key factors and provide important information for diagnosis and therapy. METHODS:Expressing of E—cad,PCNA and p53 were detected in 44 PTC and 20 normal thyroid tissues by a Labelled Streptavidin Biotin immunohistochemical method(LSAB).RESULTS:1.Expression for E—cad mainly located in cytoplasm,positive rate was 36.36% in PTC,which showed significantly lower than that in normal tissues(100%)(p<0.005).2.The positive rates of PCNA,p53 were 84.09% and 38.64% respectively in PTC which appeared significantly high than that in normal tissues(p<0.005).3.The E—cad with lower and p53 with higher frequency in lymph node metastasis PTC than that in lymph node negative(p<0.005).4.There were no relationship between the expression of E—cad in PTC and the PCNA.CONCLUSIONS:1.P53mutations,PCNA overexpression and E—cad abnormal changing may play an important role in the carcinogenesis and progression for human PTC.2.The alteration of p53,PCNA and E—cad may reflect some malignant biology of PTC,and use as a molecular index for clinical diagnosis and therapy.3.The lower detected rate of E—cad in lymph node metastasis specimens of PTC may provide the information of nodal metastasis status.4. Serial tests of E—cad,p53 and PCNA could be used as the markers to evaluate lymph node metastasis of PTC.It indicated the prognosis and provide the proper operational procedure and accessorial treatment potentially.
