Objective: the study is designed to discuss the correlation between the male rat's sexual behavior and its 5-hydroxytryptamine(5-HT) content. Methods: the animal model was produced by means of putting special moulds on the male rats being bred in cages separately. Using CHAIHUSHUGAN POWDER as trial drug to smooth the circulation of liver-Qi, the targets such as oltaction-licking latency % intromission latency, intromission frequency, ejaculation latency, ejaculation frequency, post-ejaculation interval had been observed. Fluorospectrophotometry had been used to measure the content of 5-HT in respective specimen which had been produced by separating plasm, fluid of broken platelets from rat's venous blood and homogenating the hypothalami. Results: The model groop's content of 5-HT reduced in plateleds and hypothalami while raising in plasm obviously. In copulatory tests, the model group rats had been observed morbid changes such as lengthened olfaction-licking latency and intromession latency , shortened Ejaculation latency and post-ejaculation interval. Above-mentioned targets appeared differential reversion in rats of the treated groop. Conclusions: There are material basis for influence of psychological factors in sexual function. The study has proved that there is association between the changes of sexual behavior and 5-HT in peripheral blood and center nervous system of male rats suffering liver-Qi-depression syndrome, rather, we observed the model groop's content of 5-HT reduced in platelets. From which as well as according to TCM theory, we put forward a theory foundation for treating male sexual dysfunction 'by means of regulating liver in TCM, moreover, provided a clu,e for modern studies on sexual disorder caused by depression of liver-Qi.
