| Objective:To study the mechanism relative to therapeutic effects of Matrine on adenine-induced renal interstitial fibrosis in rat. Methods :Sixty male Wistar rats were selected and randomly divided into 3 groups (1)Normal control group has 7 rats; (2)pathological group has 24 rats; (3)Matrine group has 28 rats. Each rat in pathological group and Matrine group was infused with Adenine (250mg.kg-1d-1) into stomach. Following one week, each rat in Matrinegroup was also infused with Matrine ( 200mg/l. 73m2.d-1) into stomach. Since then, two weeks, three weeks and four weeks later respectively, to determine serum urea nitrogen, creatinine, IL? and to examine pathology of kidney in pathological group and Matrine group, in which the results compared with that in normal control group. Immunohistochemical staining for TGF -β1 express and distribute in kidney, radioimmunoassay for serum IL-6 and HE staining for semiquantitative analysis of tubulointerstitial injury.Results:In Matrine treated group, the renal function and the serum IL-6 is obviously lower than that in pathological group (p<0.05, p <0.05) and TGF-β1 express level lower to compare with pathological group (p <0.05), but all these indexes is higher than normal control group (p <0. 05).After treating , in Matrine group the index of tubulointerstitial injury is lower than that in pathology group (p <0. 01) and have certain prescroption relations.Conclusion:Matrine can protect renal function and can lighten the renal tubulointerstia 1 fibrosis in adenine-induced model of rat, which were involved in suppressing level of IL-6 and the expression of TGF-β1. |
