Angiotensin Ii Receptor Antagonist Olmesartan The Synthesis

The synthetic process of a new angiotensin II receptor antagonist, Olme-sartan medoxomil was studied in this paper.The context involved in the development of antihypertensive agents about the renin-angiotensin system(RAS), summarized the advancement of angiotensin Ⅱ receptor blorkers(ARB),explained ARB physiological mechanism and commercial value,cited the better treatment in comparision between Olmesartan medoxomil with the other "saltans".The synthesis of Olmesartan medoxomil was particularly studied in this paper.The synthetic route was designed and modified so ao to adapt to industrialization.The totle yield of Olmesartan medoxomil was 35.6%,which was prepared by twelve steps (including synthesis of XI).Because of technology patent applied in China,its process must escape the patent.This paper's method do promptly it.In addition,The standard of qualified control was made under experimental conditions.The structure of Olmesartan medoxomil was confirmed by UV, IR, ¹H-NMR, ¹³C-NMR, MS, DSC.
