Objective: By studying the chronic urticaria's type of syndrom and examination on each type of chronic urticaria's Th1 and Th2 cell cytokinel, and expliciting the relation between them. To approach the pathogenesy of chronic urticaria, and provide the experimental evidence of Chinese medicine treatment of chronic urticaria. From the study in recent years, it can be seen that chronic urticaria andfuction diseqitibruum of T helper cell sudgroup has close correlation. Secretory cytokine level of Th1 cytokine decreases and level of Th2 cytokinetrends to steps up. After treatment Th1 cytokine and Th2 cytokine trends to equilibration. So the chronic urticaria patients have disequilibriumof Th1 cytokine and Th2 cytokine. We have not retrieve the study about chronic urticaria's type of syndrome and diseqilibruum of T helper cell sudgroup.Method:1 selective standard of chronic urticaria patients.1.1 The surface of skin and mucous memhrance occueerd recurrent xheals and theduration was not beyond twenty-four hours.1.2 The incidence could not be under two times weekly.1.3 The progress was over six weeks.1.4 The antihistamine drugs was not used in one week and the steroid hormone and the immunosupppression drug were not uesd in one month. 2 Exclusive standard of chronic urticaria patients2.1 The physical urticatia was excluding because of the coldness and the pressure and so on.2.2 The autoimmunitydisease was excluding.3 The standard of syndrome differentiation3. 1 Syndrome of exterior attacked by wind-heat: the color of wheal is bright red, the patient felt hot and very itching, to accompany with ferveseence, nephrasthenia asthma, Swelling of throat, the skin rash will aggravate when the patient in the hot environment, thin lingual fur and white or yellow, pulses is floating and quick.3.2 Syndrome of exterior tightened by wind-cold Syndrome of exterior attacked by wind-heat: the color of wheal is white, the skin rash will aggravate when the patient meet wind - cold and anesis in the hot environment, hydroadipsia. texture of tongue is light, coated tongue is white, pulses is floating and tight.3.3 Syndrome of hemo penia wind dryness: recurrent attacks, protraction for long time, the skin rash will aggravate in the post meridiem or at night, to accompany with upset and tantrum, dry mouth, feverish sensation over the palm and sole, red tongue hyp- bodyfluid, pulses is deep and trifling.4 Monocytic seperated technique in venous blood: there are 68 petients of chronic ueticaria and fresh venousblood was going to be drawn from every patients. In order to obtan Lymphoeytes and Monocyts the Ficoll iso-sedimentation seperation was used and the cell susqension was produced whit RMPI1640 and the celluar tele-concengtration was 1×10~6/ml.5 Detdcting method of cellular vital force: the teypan blue execlusion test wasselected and after stanging with 0.4% trypan blue counted vital cell could be above 95% in three minutes.6 Routine obseration of cultured cell.6.1 The changes of form and tiny formation: the volum of lymphoblasts largened and the nuclei were transparent and ball agglutiphenomenon occurrded.6.2 Observation of culture medium: the normal fresh culture medium containing phenolsulfonphthalein was pewchblow and as so as it became primary colors and the possibility of bacteria contamination needed be attented.7 The cuitivation of pro-generation cell: the cells were coltured in vitro with RPMI1640 culture medium in the 96 cell culture plates and had been hatched for 48 hours in the carbondioxid incubator on the condition of 37℃, and then had been centrifugated by mesns of 2000r/min, at last the surperior-schedul liquor were collected and were preserved on the condition of -20℃.8 The drtecting method: the IL-4 and IFN-γcontents from the surerior-schedul liquor of every group were detected by means of ELISA.Result:1 The change to the form and the tiny constitution: after being stimulated by the PHA, T cells occoured a series of proliferation response as cells volum largening and the nucleus distinctnees and cyloplasmic manifold and the vacuole occurred and the chromatin rarefaction and the ball coagulation phenomenon, all of this proved that the lymphocyte had translated into the lymphocytoblast.2 After being stimulated by the PAH, To detect the leveI of L-4 cytikine about chronic urticaria's PBMC, it can be seen that the type of Syndrome of exterior attacked by wind-heat, Syndrome of exterior tightened by wind-cold, Syndrome of exterior attacked by wind-heat, Syndrome of hemo penia wind dryness are higher than health adult, it has statistical significance. The tL-4 level of syndrome of hemo penia wind dryness group is higher than the other gpoups, but it has't statistical significance. To detect the leveI of IFN-γcytikine about chronic urticaria's PBMC, it can be seen that the type of Syndrome of exterior attacked by wind-heat, Syndrome of exterior tightened by wind-cold, Syndrome of exterior attacked by wind-heat, Syndrome of hemo penia wind dryness are lower than health adult, it has statistical significance. The IL-4 level of syndrome of hemo penia wind dryness group is lower than the other gpoups, but it has't statistical significance.Conclution:1 The patients of CU has exist Th1/Th2 cytokine disequilibrium. Different type of syndrome about CO 's IL-4 cytokine are higher than health adult and IFN- are lower than health adult.2 Chronic urticaria's type of syndrom can not based on the level of Th1/Th2 cytokine.3 We can study the effection that traditional chinese medicine and prescription treat on diferent CU's type of syndrome cytokine, and approach mechanisms about traditional chinese medicine and prescription ettect on CU.
