Xiao Chai Hu Tang And Its Prevention And Treatment Of Drug Group Compatibility Hyperlipidemic Fatty Liver Of Experimental Research

The article were divided to two parts,literature overview summarize clinic experimental study of xiao chaihu tang (XCHT). Experimental study was divided to following two parts.Experiment 1Aim:to study the effect of XCHT on the hyperlipidemia and hyperlipidemic fatty liver animal model and to observe the change of the model in five weeks.Methods:the rats were divided into five groups randomly,control 30,model 30, high-dose group of XCHT 10, low-dose group of XCHT 10, Simvastatin group 10. In the fist week, the rats of model,XCHT, Simvastatin groucontrolclysterized with fat emulsion( 10 ml·kg - 1),while the rats of control group were clysterized with water( 10 ml·kg - 1) in the morning every day, all rats were fed with a standard diet.In the 8th day, 10 rats were taken randomly from control group and model group, the materials were drawn, the serum TG, TC, LDL-C,HDL-C were detected.From the 8th day, the rats of model, XCHT, Simvastatin group were clysterized with fat emulsion( 10 ml·kg - 1),the rats of control group were clysterized with water( 10 ml·kg - 1) in the morning every day. The herb groups was clysterized with Simvastatin 4.67mg/kg and XCHT (18.9g/kg,9.45 g/kg ) respectively in the afternoon every day;the rats of control group and model group were clysterized with water( 10 ml·kg - 1) in the afternoon every day.In the 15th day, 10 rats were taken randomly from control group and model group, XCHT groups,Simvastatin group,the materials were drawn, the serum TG, TC, LDL-C,HDL-C were detected, All groups deal with as above. In the 22th day, 10 rats were taken randomly from control group and model group, XCHT groups,Simvastatin group,the materials were drawn, the serum TG, TC, LDL-C,HDL-C were detected,the liver and body were weighed, the coefficient of liver were also Calculated (the coefficient of liver=weight of liver/ weight of body×100%).From the 22th day, all rats were fed with a standard diet, spontaneous recovery were observed in 14 days.In the 29th day, 10 rats were taken randomly from control group and model group, the materials were drawn, the serum TG, TC, LDL-C,HDL-C were detected,the liver and body were weighed, the coefficient of liver were also Calculated. In the 36th day,all groups deal with as above.Results:At the end of the fist week,compared with control group,during making model,model group TG,HDL-C is higher than control group(P<0.05).At the end of the second week , compared with control group,during making model,model group TG,TC,HDL-C is higher than control group(P<0.05). compared with model group,the serum TG of the XCHT groups,Simvastatin group group rats were markedly lower(P<0.05). At the end of the third week, compared with control group,model group TC,TG,LDL-C, HDL-C is higher than control group(P<0.05);compared with model group,the serum TG, TC of the XCHT groups,Simvastatin group group rats were markedly lower(P<0.05);compared with model group, the coefficient of liver of high-dose group of XCHT group,Simvastatin group,control group rats were markedly lower(P<0.05), the weight of the XCHT groups,Simvastatin group, control group were not different with model group.At the end of the fourth week, compared with control group, the coefficient of liver of model group were markedly higher(P<0.05), the serum TG,TC,LDL-C,HDL- C of model group rats were not different.At the end of the fifth week, the coefficient of liver, the serum TG,TC,LDL-C,HDL- C of model group rats were not different with control group.Experiment 2Aim:to study the effect of separated paired drugs in Xiaochaihu tang(XCHT) on the hyperlipidemia and hyperlipidemic fatty liver animal model.Methods:The components of XCHT were divided into Chaihu-Huangqi group, A group, Banxia-ganjiang group (B group) Rensheng-Gancao-Dazao group (C group) according to their functions.Then they were regrouped on the basis of the orthogonal test table of L8(27), there were eight groups: A group,B group,C group,A+B group(AB), B+C group(BC), A+C group(AC),A+B+C group.The rats were divided into nine groups randomly,control group,model group, A group,B group,C group, AB group, BC group ,AC group and ABC group,there were 9 rats every group .In the fist week,the rats of model group, A group,B group,C group, AB group, BC group ,AC group and ABC group,were clysterized with fat emulsion( 10ml/kg ),while the rats of control group were clysterized with water( 10ml/kg) in the morning every day, all rats were fed with a standard diet.From the 8th day,the rats of model group, A group,B group,C group, AB group, BC group ,AC group and ABC group,were clysterized with fat emulsion( 10ml/kg),while the rats of control group were clysterized with water( 10ml/kg) in the morning every day.The rats of AB group were clysterized with AB tang(11.9g/kg),the rats of AC group were clysterized with AC tang(14.7g/kg), the rats of BC group were clysterized with BC tang(11.2g/kg), A group were clysterized with Atang(7.7g/kg),B group were clysterized with B tang(4.2g/kg) ,C group were clysterized with C tang(7g/kg), ABC group were clysterized with ABC tang(18.9g/kg) ,respectively in the afternoon every day, the rats of control group and model group were clysterized with water( 10ml/kg) in the afternoon every day, all rats were fed with a standard diet.In the 22th day, all rats were executed and the materials were drawn, the serum TG, TC, LDL-C,HDL-C and the hepatic TG,TC were detected,the liver and body were weighed, the coefficient of liver were also Calculated (the coefficient of liver=weight of liver/ weight of body×100%).Results:All groups had decreased markedly the level of the serum TG(P<0.05), all groups had decreased the level of the serum TC,LDL, B and C group had markedly effect on the level of the serum TC,LDL(P<0.05). All groups had no markedly effect on the level of the serum HDL-C. All groups had decreased the level of the hepatic TG,TC, A,B,C,AB groups had markedly effect on the level of the hepatic TG, A,Bgroups had markedly effect on the level of the hepatic TC。Conclusion:The experimental animal model of hyperlipidemia and hyperlipidemic fatty liver in rats were successfully established for 1 week and 3 weeks fed with fat emulsion. After one week of spontaneous recovery, compared with control group, the coefficient of liver of model group were markedly higher, the serum TG,TC,LDL-C,HDL-C of model group rats were not different. After two weeks of spontaneous recovery,all index of model group rats were not different with control group markedly. XCHT have decreasing effect on the level of the hepatic TG,TC, the coefficient of liver, the serum TG,TC.All groups have decreasing effect on the level of the hepatic TG,TC, the coefficient of liver and the level of the serum TG, TC,LDL, had no markedly effect on the level of the serum HDL-C.In this topic, we found that XCHT and separated paired drugs in XCHT have decreasing effect on the hyperlipidemia and hyperlipidemic fatty liver animal model, the different separated paired drugs in XCHT have the different effect on the different index. It provides the way to study new drug.
