Flos Carthami is the drying flower of Carthamus tinctorius L.which is 1-year-span herb of family Compositae.In China,it is mainly produced in Xinjiang,Sichuan, Yunnan,Henan,Hebei,Shandong,Zhejiang,Jiangsu and etc.Traditional effect of Flos Carthami is promoting blood flow,and it is usually used to treat cerebrovascular diseases,cardiovascular diseases and gynaecological diseases and so on.Major constituents of Flos Carthami are pigments,flavonoids and phenolic acid.At present the research of Flos Carthami quality is mainly focused on the following two aspects, i.e.the fingerprint spectrum study and the quantitative analysis based on one component.On the foundation of previous research in field,the quality of Flos Carthami was analyzed and evaluated via fingerprint spectrum analysis and multi-ingredient content determination to 32 batches samples from main production areas with methods of HPLC-electrochemistry examination,the UV diode array examination technology and the UV spectrophotometer.Furthermore,extraction process of effective parts of Flos Carthami was also studied.Specifically as follows:1.The study on Flos Carthami fingerprint and content determination of Hydroxysaffiower yellow A.Using HPLC coupled with electrochemical detector, UV diode array detector,and mass spectrometry detector,a method was established to determine the content of hydroxysaffior yellow A and fingerprint analysis of Flos Carthami.21 common chromatographic peaks were selected as characteristic peaks in the chromatogram of sample solution of Flos Carthami.7 peaks were identified as the peaks of hydroxysafflor yellow A,6-hydroxykaempferol-3-O-Glucoside,rutin, quercetin-3-O-glucoside,kaempferol-3-O-rutinoside,quercetin,kaempferol.The result showed that the samples existed differences.The relative peak areas of common peaks were taken as variables,and were analyzed by SPSS.The samples of Xinjiang, Yunnan provinces were approximately classified by region as a result.The content of hydroxysafflor yellow A was from 0.35%to 3.58%. 2.The study on the determination of adenosine and cartharmin.Concentration of adenosine and cartharmin were determined by HPLC-UV and UV spectrophotometer, respectively.In 32 batches of samples,the content of adenosine existed the big differences,in 0.03‰~0.49‰between.And the absorbance of cartharmin was from 0.202 to 0.634.3.The study on extraction process of antioxidative part of Flos Carthami and its quality standards.According to the pharmacological results,the water-extracted part was settled as the effective part of Flos Carthami.Through the orthogonal design, the water extraction process was established,and a purified process by macroporous resin had also been set up.Meanwhile in the paper the quality standards of the antioxidative part of Flos Carthami were established and drafted.
