Rhizoma alismatis is the dried tuber of Alismaceae plant Alisma orientalis (Sam.)Juzep.,which is being recorded in China pharmacopoeia.The study using the column chromatography technology to separate and purify the effective components of reducing blood lipid in Rhizoma alismatis,5 compounds had been isolated,which purity are above 95%(area normalization method)by using thin layer chromatography and high performance liquid chromatography to testing their purity,fitting the requirement of structure identification;5 compounds had been identified asβ-sitosterol,alismol,alisol B-23-acetate and alisma Orientale C-23- acetate,emodin; investigating the stability of alismol by using thin layer chromatography and high performance liquid chromatography,result indicates alismol is stability during 6 moths by preserved in the condition of room temperature and away from light.We set up the quality criteria of Rhizoma alismatis.Testing moisture content,heavy metals,arsenum,ash content and acid-insoluble ash of Rhizoma alismatis from 10 places of produce;testing the ethanol-soluble extraction,the average content is 12.77%,we suggest the content of ethanol-soluble extraction in Rhizoma alismatis should be above 10 %;we take alismol as reference substance to set up the thin layer chromatography method of Rhizoma alismatis,alismol are tested in 10 samples;we set up the assaying method of alismol by using high performance liquid chromatography,alismol has good linear correlation within the range of 4.9580～19.8320μg,average recovery is 99.66%,RSD is 1.18%,the method is convenient,sensitive,accurate and suitable for the quality control of Rhizoma alismatis;testing the content of alismol in 10 samples,the average content is 0.173mg/g,then we suggest the content of alismol in Rhizoma alismatis should be above 0.014%.In this paper,we separated and purified the effective components of reducing blood lipid in Rhizoma alismatis,getting the effective component of Rhizoma alismatis,taking the effective component as reference substance to set up the quality control method which could reflect the function and clinical application of Rhizoma alismatis,proving current standard,establish the foundation for the quality control of Rhizoma alismatis and its compound preparation.
