| Objective:To study the effect of Yifu-Juanbi granule on the pathomorphology of the synovial membrane of adjuvant arthritis rats after observing the morphologic changes and the activity of NF-κB,FasL,TNF-αand Caspase-3 in rats.Methods:The experimen was performed on 60(Sprague-Ddawley) SD rats which were separated into six groups--normal group,model group,high-dose group,middle-dose group and low-dose group.All the anklebones of the rats with adjuvant except those of normal group were injected.After 14 days,all the rats were given with corresponding medicine until the ending of our experiment.Then the pathomorphology of the synovial membrane of adjuvant arthritis rats was detected by microscope.And NF-κB,TNF-αand Caspase-3 were tested.Results:Invasion and hyperplasia of the inflame,atory cells in the synovial membrane of the adjuvant arthritis rats were obvious,while the pathological changes of the Yifu-Juanbi granule groups was lightened than the model group obviously(P<0.05).The change of the control group's histomorphology was clear.,with the incrassation of synovial lining cells,the infiltration of Inflammatory cells and the erosion of nephelium of blood vessels on bones.However,the changes of the Yifu-Juanbi granule groups were lightened obviously.The level of TNF-α,NF-κB and Caspase-3 into model group increased,compare with normal group,showed significant different(P<0.01~0.05).In high--dose group,middle-dose group and low-dose group,the level of TNF-αand NF-κB obviously decreased,Caspase-3 increased obviously,compared with model group,showed significant different(P<0.01~0.05).Conclusion:Yifu-Juanbi granule can alleviate the change of the pathomorphology affected by rheumatoid arthritis.The mechanisms might relate to reduce TNF-α,NF-κB level and impove Caspase-3 levels so as to inhibit the apoptosis. |
