Xiaohongshen is the dry root and rhizome of Rubia.yunnanensis(Franch.) Diels,which is widely spreaded in Yunnan province and Guizhou province.Rubia yunnanensis is used by multi-nationalities(the Yi nationality,the Pai nationality,the Pumi nationality,the Lisu nationality race).It has enriching the blood and promoting blood flow,expelling wind and eliminates the wet effect.About its quality standard,there are only origin,character identification,the Microscopic identification,Chemical identification and so on in Chinese Pharmacopoeia edition 1977,Yunnan Province Drugs Standard edition 1974,Guizhou Province Traditional Chinese Medicine Quality Standard edition 1988,Guizhou Province Traditional Chinese Medicine,Ethnomedicine Quality Standard edition 2003.The studies on Rubia.yunnanensis quality have not been reported.The TLC qualitative method,total anthraquinone Spectrophotometric Determination of quantitative method and quantitative measurement method are studied and established. The results fill the gaps in the research of Rubia.yunnanensis quality standard.The results are as follows:The standard reference substance used in the quality evaluation are separated a nd identified for the first time,and the purity,the structure and the stability had be en studied,which conformed to the New medicine researched technical guidance prin ciple of Traditional Chinese Medicine.Two compounds A and B are geted from th e acetidin site.A is 1,4-naphthohydroquione -4-O-β-D-glucosid e;Bis2-methyl-1,3,6-trih ydroxy-9,10-anthraquinone-3-O-(O-6'-acetyl)-α-L-rhamnosyl(1â†'2)-β-D-glucoside.Among them,compound A is obtained for the first time from this plant;compound B is th e main active constituent of anti-heart muscle ischemia.The quantitative analytical method of total anthraquinone is established:Compound B is as the reference substance to set up the Visible Spectrophotometry assaying method of total anthraquinone in Rubia.yunnanensis for the first time,which could be taken as the quality control index.Five different sourced samples are determined using the method,the results are between 0.62%and 0.78%.The quantitative analytical method of compound B is established:Compound B is as the reference substance to set up the RP-HPLC assaying method of compound B in Rubia.yunnanensis for the first time,which could be taken as the quality control index. Five different sourced samples are determineed using the method,the results are between 0.37%and 0.48%.The qualitative analytical method of compound A is established:Compound A is as the reference substance to set up the TLC assaying method of compound A in Rubia.yunnanensis for the first time.Five different sourced samples are distinguished using the method.Compound A can be taken the TLC index.Five batches of medicines for the moisture,the total ash and the acid insoluble ash and the ethanol-soluble extractives were determined to participate for as Rubia.yunnanensis quality evaluation.
