Protection Effects of Ulinastatin against Lung Warm Ischemia-Reperfusion Injury in RatsPurpose:To investigate the protective effect of Ulinastatin against lung ischemia-reperfusion injury in rats and compare the contribution of two different doses.Methods:Thirty SD rats were randomly divided into three groups. In Ischemia-Reperfusion group (IR group, n=10),the hilum of left lung was blocked for 1 hour, before it was reopened. After 2 hours of reperfusion, blood-gas analysis was measured for left lung venous blood before the rats were killed.Upper parts of left lungs were assayed for wet/dry weight ratio (W/D), middle parts were taken for microscopic study, other parts were frozen in liquid nitrogen for detection of myeloperoxidase concentration(MPO); In Ulinastatin Conventional dose group (UC group, n=10) or Ulinastatin High-dose group (UH group, n=10), rats received 10000 U/Kg or 20000 U/Kg Ulinastatin intravenously before blockade of the left hilum, and other procedures were same as IR group.Results:IR group was compared with the UH group, there was statistically significance in PO2 of blood gas analysis and W/D [(109.5±18.3) mmHg vs (128.5±17.7) mmHg,5.30±0.13 vs 5.08±0.25, P<0.05];For the results of MPO concentration, among IR group, UC group and UH group, there was significant difference between each two groups [(57.36±10.51) ng/mL, (45.74±8.13) ng/mL and (28.30±6.28) ng/mL, P<0.05]; Tissue slice (HE stain) showed more inflammatory cells infiltration in IR group.Conclusions:This study demonstrated that Ulinastatin has a protective effect on rat lung warm ischemia-reperfusion injury, and there was dose-dependent. The Experimental Model of Orthotopic Left Allograft Lung Transplantation in Rats without Using CuffPurpose:To establish a stable model of rat orthotopic left lung transplantation using direct sutureing of vessels and bronchi.Materials and Methods:Ten SD rats weighed 250-350g were used as lung donors and recipients respectively. Airway and pulmonary vessels were reconstructed microsurgically using continuous running suture technique. Survival time were recorded and donor lungs were checked by autopsy.Results:All 10 rats received left lung transplantation were weaned from ventilator successfully. Both the cold ischemia time and warm ischemia time were about 40 minutes. The total procedure took approximately 130 minutes. Autopsy was used to check the patency of anastomosis. No thrombosis or air leak was found.Conclusions:Direct microsurgical suture can be used to establish an experimental model of orthotopic left allograft lung transplantation in rats. This method proved to be stable, reliable and is similar to that used in clinical practices.
