Purpose:Ootheca mantidis is a Traditional Chinese medicine listed in Chinese Pharmacopoeia 2005. As a kidney toning and spontaneous emission arresting agent, Ootheca mantidis it is widely used for treatment of childhood nocturnal enuresis and urinary incontinence. Based on our previous research on Ootheca mantidis,we sought to investigate the antidiuresis effect, chemical components, salt-processing techniques, as well as quality standard of Tuan Piaoxiao.Material and method:Material: Ootheca mantidis is provided by Benxi of Liaoning Province.It is been verified by Professor Zhai YanJun(Liaoning University of Traditional Chinese Medicine)as genuine dried egg capsule of Tennodera sinensis Saussuer.It is been named as Tuan Piaoxiao by customs.Method:1. Utilizing water load rat model,we compare the antidiuresis effect of unprocessed, processed Ootheca mantidis and different portion of extraction layers separated by multi-solution extraction by measure the urine volume/body weight rate,0-5 hour after administration.2. Using GC-MS,TLC,and HPLC to compare the chemical component of Ootheca mantidis, before and after the processing.Sepatate the active ingredient of Ootheca mantidis by silica column chromatography,gel column chromatography and recrystallization.3. Choosing Tyrosine and 1 - (3,4 -dihydroxyphenyl) -2 - acetamido– ethanone as quality control indicator,using HPLC and variance analysis to optimizing the salt stir-frying technique.4. The quality criteria of Ootheca mantidis was established by TLC and HPLC.Results:1. While using the same source material, cutting technique, and rat model, we compare both acute toxicity and urine volume effect on water load rat and control group. Compare to control group, the rat shows reduced urine volume after underwent treatment with unprocessed and processed Ootheca mantidis for 15 days, while the most significant urine volume reducing was observed 0-1, and 1-2 hour after treatment. The volume reducing rate after 0-1 hour of treatment by unprocessed, steam stir-frying, and salt stir-frying Ootheca mantidis was 41.12%, 69.76%, and 65.11%, respectively.Study the antidiuresis effect from different portion of extraction layers separated by multi-solution extraction. Results shows that the water layer and butanol layer contain active ingredient, which decreased urine volume by 69.95% and 64.22%, when compare to control group.2. We utilized GC-MS,TLC, and HPLC to compare the chemical component of Ootheca mantidis, before and after the processing. While there was no new absorption peak after process, the quantity of active ingredient changed. We able to identify nine components from the butanol layer part of extraction. They are: 1 - (3,4 -dihydroxyphenyl) -2 - acetamido– ethanone,Tyrosine, cyanuric acid, uracil, hydroquinone, 2,2'-dicarboxylic acid, pileric acid, 3-aminopiperazine-2,5-dione,β-sitosterol. Total nine lipid components were identified for the first time.3. The salt stir-frying technique was studied by vary salt quantity, smooth time, frying temperature, and frying time, using Tyrosine and 1 - (3,4 -dihydroxyphenyl) -2 - acetamido– ethanone as indicator, L9(34) orthogonal for variance analysis. The result show that frying time is critical to the concentration of 1 - (3,4 -dihydroxyphenyl) -2 - acetamido - ethanone,1,5 - bis (p-hydroxyphenyl) -2,5 - diamino -3,4 - dihydroxy-hexane. The optimized processing technique is: add 30L 8.3% NaCl water solution for every 100kg raw material, smooth for one hour, then frying for 10'at 100 oC.4. We choose Tyrosine and 1 - (3,4 -dihydroxyphenyl) -2 - acetamido– ethanone as indicator to study the quality of unprocessed and processed Ootheca mantidis. The preliminary quality standard was established by measuring water, total ash, and acid-insoluble ash, in unprocessed and processed Ootheca mantidis from fifteen origins. Conclusion:1. Compare to unprocessed Ootheca mantidis, the antidiuresis effect is significantly stronger after salt stir-frying.Its reduce urine volume (0-1 hour after administration) by 69.76% in rat which been treated for 15 days.2. The water layer and butanol layer of muti-solution extraction contain active ingredient of Ootheca mantidis.3. The optimized salt stir-frying procedure is add 30L 8.3% NaCl water solution for every 100kg raw material, smooth for one hour, then frying for 10'at 100 oC.4. According to the experimental result, the moisture in Ootheca mantidis should be less than 13%,the total ashes content with-in 10%, ashes imdissolvable in acid with-in 1%, water extracts no less than 9%, ethanol extracts no less than 11%, Tyrosine no less than 0.1494%,1 - (3,4 -dihydroxyphenyl) -2 - acetamido - ethanone no less than 0.0045%. This standard provides the scientific basis for the quality evaluation of Ootheca mantidis.
