With the wide use of new immunosuppressive matches and the constant improvement of the technology, Renal transplantation of acute rejection has been brought under eff-ective control,but long-term survival of the graft is still no obviously improved.In the world the severe increasingly scarce organ source situation, how to make the recipie-nts hard-won long-term graft attrition, become an important topic and urgent task for the transplant workers.However, currently the clinical transplant workers routinely offer recipients immunosuppressive which is adopted by weight, make occurred infe-ction and rejection phenomena often appear.The serious phenomena restricts the incincreasing survival of postoperative recipients.Infection has always been major complication of the renal transplantation,in transpla-ntation approximately 75% of patients have occurred at least once infection. In any st-age of postoperative infection could lead to death of the patients. How to effectively control the infection protect renal function, reduce rejection, improve recipients surial become clinicians urgently solved the difficulty. So the study of infection in the path-ogenesis of characteristics and adopt effective prevention and treatment measures are all very important. And neutrophils as nonspecific an important part of the immune sy stem, involve in the body's resistance to infection and adjust the immune physiologi-cal processes. Ncutrophils adhesion molecules GD11b/CD18 is integrin family beta 2 and a member of the family, mainly express in neutrophils and mononuclear cells, through with endothelial cell adhesion molecules combined with endothelial cells which mediated leukocytes interaction. The expression level of CD11b/CD 18 in neutrophils'surface is infla- mmation key factors,which can be used as cell adhension function indices. In recent years, the research of CD11b/CD18 has been extensively reported, and many research use flow cytometric by perioperative blood to test CD11b/CD18, but the detection of intracelluar CD11b/CD18 mRNA expression level of renal transplantion recipients has never been reported.Myeloperoxidase (MPO) is the secretion peroxidase of activated neutrophils, monocytes and macrophages. MPO generate hypochlorite which has strong antimicrobial activity. MPO is the important enzymes in the body's defence function, and is the important composition which dependent on Oxygen to sterilizate of neutrophils and mononuclear cells. The current research of MPO focuses on the cardiovascular diseases, while the detection of MPO in renal transplantation recipients in peripheral blood neutrophils rarely has been reported.Objective:To study the expression level of adhesion molecules CD11b/CD18 and the activityof myeloperoxidase (MPO) in the peripheral blood neutrophils of renal transplant recipients, the correlation of CD11b/CD18 and MPO in renal transplant recipients.To discusses CD11b/CD 18 and MPO the significance of pathogenesis in infection patients with renal transplantation.Methods:1.By density gradient centrifugation separate renal transplantation patients and healthy controls peripheral neutrophils.2.Reverse transcription-polymerase chain reaction (RT-PCR) was applied to detected CD11b/ CD18 mRNA expression in peripheral blood neutrophils cells (PMNs)in renal transplant patients.3. Flow cytometry was used to detect neutrophils adhesion molecules CD11b/CD18 expression level.4.Spectrophotometer colorimetric method was used to detect MPO expression of peripheral neutrophils.5. SPSS 17.0 statistical software was applied for the data statistics. x±s and one-way ANOVA(F test) were used to compare the data. The analysis of Bonferroniwas used to compaer these data. Relevance with linear related analysis Pearson was used to compaer these data.Test level is a=0.05.Results:1.Renal transplantation stable group, infection group of peripheral neutrophils CD11b/CD18 the mRNA expression level for0.495±0.14,0.417±0.11,,healthy controls peripheral blood neutrophils CD11b/CD 18 mRNA the expression level for the 0.528±0.12.Stable group and infection group, infection group and controls results are statistically significant different(P<0.05). The differences between controls and postoperative stable group were not statistically significant (P>0.05).2. Renal transplantation stable group, infection group CD11b/CD18 molecular expression level in neutrophils peripheral blood were25.43±1.52,22.55±1.71, controls molecular expression of CD11b/CD 18 in peripheral blood neutrophils were 28.04±1.52.Stable group and infection group, infection group and controls results are statistically significant different (P<0.05). The differences between controls and postoperative stable group were not statistically significant (P>0.05)3. Renal transplantation patient stable group, infection group activity of MPO in peripheral blood neutrophils were respectively65.81±4.91,54.32±2.00, controls was 66.78±2.80. Stable group and infection group, infection group and controls results are statistically significant different (P<0.05). The differences between controls and postoperative stable group were not statistically (P>0.05).Conclusions:1. When the infection occurred after kidney transplantation, CD11b/CD18mRNA and molecular expression level and the activity of MPO in peripheral neutrophils were significantly decreased.2.CD11b/CD18mRNA and molecular expression level and the activity of MPO for the kidney transplantation patients exist positive linear correlation.
