Objective:To study the role of AQP1 in the development of asthma and provide new direction of prevention and treatment for asthma,by observing the distribution and expression of AQP1 and effect of jin-shui-bao and dexamethasone on expression of AQP1 in The acute bronchial asthma lung tissue.MeThods:SD rats were randomly divided into A,B,C,Dgroup (each group contained 8rats), including group A (saline control group), B,C,D groups were asthma group, jin-shui-bao interention group (gastric lavage 4g/kg), dexamethasone interention group (intraperitoneal injection 2.5 mg/kg)in turn, The intervention groups were done in the excited phase, and rats were killed after 2 weeks. Observation changes of lung tissue;Counting and classification inflammatory cell in bronchoalveolar lavage fluid (BALF);Detecting IL-4, INF-γchange by ELISA; Observation The distribution and expression of AQP1 in asThmatic rats lung tissue by immunohistochemistry and western blot.Results:①It can see obviously diffuse exudative inflammatory changes in asthmatic lung tissues but There were lesser degree of inflammatory changes in Jin-Shui-Bao,DM intervention groups.②Eosinophils,neutrophils,lymphocytes,IL-4 were significantly higher in asthmatic control group Than That of saline control group, but The INF-γwere significantly lower Than saline control group; Total cells,eosinophils,neutrophils,lymphocytes and IL-4 were significantly decreased after the intervention of Jin-Shui-Bao and DM, and there were no significant differences between Jin-Shui-Bao and DM interention group.③AQP1 is mainly distributed around The alveolar capillary endothelial cells, lamina propria of The vascular endothelial cells, airway epithelial cells, lymphocytes and eosinophils; AQP1 was weaker expression in the cordyceps and dexamethasone interention group compared with asthmatic group which was strongly positive, but stronger than the saline control group (P<0.05), and there was no significant difference between Jin-Shui-Bao and DM interention group (P> 0.05).Conclusions:①AQP1 was strongly positive expression in the lung tissue of acute bronchial asthma and inflammatory cells.②jin-shui-bao and DM can inhibit inflammatory cell accumulation in The lungs together, and inhibited the expression of AQP1 significantly.③The decreassion expression of AQP1 likely to reduce inflammation cells aggregation in asthmatic lung organizationr.
