Objective; To explore the distribution of the K469E and the G241R single nucleotide polymorphism of the Intercellular Adhesion Molecule-1 (ICAM-1) Gene in Xinjiang Kazak people,and analyze the correlation between ICAM-1 gene polymorphism and cerebral infarction (CI),the possible susceptible marker of CI in Kazakan was determined.Methods: In 100 cases of cerebral infarction patients and 110 healthy controls in Xinjiang Kazak people,ICAM-1 K469E and G241R genotypes were detected by Polymerase chain reaction - restricted fragments length polymorphism (PCR- RFLP), The x2 test was adopted to check the difference in genotype distribution between cases and controlsand the accordance with Hardy-Weinberg equilibrium test for all samples.Results:1. The frequencies of genotypes of K469E polymorphism were KK 0.65,KE 0.23, EE 0.12 in the cerebral infarction group and the corresponding frequencies distribution were KK 0.473, KE 0.391,EE 0.136 in the control group, the genotypes distribution showed significant difference between two groups (x2=7.379,P=0.025).KK genotype increased greatly in the cerebral infarction group (x2=6.672,P=0.010).2.The K and E allele frequencies were 0.765 and 0.235 respectively in cerebral infarction group and 0.668 and 0.332 in the control group respectively and the frequencies of K allele was higher in cerebral infarction group (X2=4.812,P=0.028) The odds ratios (OR) was 1.617 with 95% confidence intervals (95% CI) (1.051~2.487).3. Two genotypes GG and GR of ICMA-1 G241R polymorphisms were found in xinjiang Kazaka people. The genotypes frequencies of G241R polymorphism were GG 0.90, GR 0.10,in the cerebral infarction group and the corresponding frequencies distribution were GG 0.9223,GR0.0727 in the control group respectively,the two groups were no significant difference in genotype distribution (x2=0.497,P=0.481).4.The G and R allele frequencies were 0.950,0.050 respectively in cerebral infarction group and 0.9636 and 0.0363 in the control group individually,the two groups showed less significant difference in allele frequencies.Conclusions: K469E polymorphism of ICAM-1 gene may relate to the essential cerebral infarction of Kazak in Xinjiang, and the Kazakans in Xinjiang carrying KK genotype are apt to suffer cerebral infarction.The K allele may be one of a genetic susceptibility factors for the cerebral infarction of Kazak in Xinjiang.But no evidences support that G241R polymorphism of ICAM-1 gene is associated with cerebral infarction of Kazak in Xinjiang.
