Objective To investigate the effect of different concentrations of cuscuta on sperm membrene and acrosin activity of patients with asthenospermia in vitro , analysizing the mechanism of cuscuta for asthenospermia.Methods 60 patients with asthenospermia were collected in the center for reproductive medicine of the the General Hospital of Ningxia Medical University. And their semen were incubated to different concentrations of cuscuta (5%,10%,15%,20%,25%)by coincubation, quinn's1020 fluid as control group,to analyze human sperm motility, sperm tail membrane function, sperm acrosin activity and sperm morphology in different times. Repeated measures analysis of variance was used spss11.5 software.Results (1) 15% Cuscuta preparation in 60min significantly increased sperm motility a grade, b grade, sperm vitality compared with the control group (P <0.01) . 15% Cuscuta preparation in 30min,60min,90min,120min were significantly higher than control group (p>0.05).(2)15% Cuscuta preparation in 60min significantly inceased sperm tail swelling rate compared with the control group and the time.(P<0.01),25% Cuscuta preparation in various times was significantly lower than the control group (p<0.01).(3) 15% Cuscuta preparation in 60min significantly inceased sperm acrosin activity,compared with the control group ,While there was lightly higher than in other times .5%,25%Cuscuta preparations were lower than the control group(allp <0.01);(4)15% Cuscuta preparation Sperm morphology was no statistical significance(p>0.05). Conclusions (1)The appropriate concentrations of Cuscuta in the effective time can significantly improve sperm movement, sperm tail membrane function and sperm acrosin activity;(2) The best priority time of Cuscuta on sperm asthenospermia in vitro is 60min;(3) Sperm motility,viability are positively correlated with sperm hypoosmotic swelling rate and sperm acrosin activity.
