Previous study has confirmed MARVELD1(MARVEL domain containing 1) as a candidate tumor suppressor gene, which is located on the long arm of human chromosome 10 with zone 4 sub-zone 2 (10q24.2). Its mRNA of 3238 bp contains two extrons and expresses a 18.9 kD protein, which will inhibit tumor proliferation and arrest cell cycle. As previous study on MARVELD1 predicts it may interact with histones and modified forms, we take A549 and HepG2 cell lines that overexpress MARVELD1 for the co-location analysis between MARVELD1 and histones then the results demonstrate the co-location between MARVELD1 and Histone H3.1, H4, mono-methylated H4 K20 and tri-methylated H4 K20 in mitosis prophase. We also set up pGex-4T-2/MARVELD1 recombinant and get GST/ MARVELD1 fusion protein. Then GST pull-down analysis show that MARVELD1 interacts Histone H4 K20 me1 but others. Subsequently, co-immunoprecipitation analysis also supports that.Moreover, western blotting is taken to measure the expression level of histones regulated by MARVELD1. On account of MARVELD1 overexpression, the expression level of unmodified Histone H4 and tri-methylated H4 K20 is up-regulated while the expression level of acetylated Histone H3 K9/14 is down-regulated as well as mono-methylated H4 K20.As is known mono-methylated H4 K20 plays role in cell cycle progression, genomic stability, chromatin condensation, formation of heterochromatin, gene transcription regulation and X chromosome inactivation., we consider MARVELD1 may associate with these functions and that will contribute to the farther study of MARVELD1...
