Chapter1:The food safety issues were briefly discussed, three types of harmful chemical substances studied in this thesis and their limit standards in different nations were presented, the research progress of immunoaffinity purification technology in this field were introduced, on the basis of these reviews, our own research contents and the innovative points were put forward.Chapter2:A method was developed for simultaneous determination of10kinds of mycotoxins in6kinds of matrix of rice, wheat kernel, corn, peanut, vinegar and sauce by immunoaffinity chromatography coupled with ultrahigh performance liquid chromatography tandem mass spectrometry (IAC-UHPLC-MS/MS). The sensitivity and accuracy of the method were improved by optimizing the conditions of MS acquisition parameters and mobile phase and selecting the best injection solution. The samples were cleaned by the Myco6inlTM immunoaffinity column, which led the better purification effect and the lower limits of detection ranging from0.2to20μg L-1, the mean recoveries were between74%and105%. The developed method was applied to the detection of mycotoxins in actual commercial agricultural products.Chapter3:A method was developed for simultaneous determination of12kinds of sulfonamides in4kinds of matrix of pork, beef, milk and egg by immunoaffinity chromatography coupled with ultrahigh performance liquid chromatography tandem mass spectrometry(IAC-UHPLC-MS/MS).The sensitivity and accuracy of the method were improved by optimizing the conditions of mobile phase and immunoaffinity purification, selecting the best injection solution and evaluating the matrix effect. The samples were cleaned by the IAC-SULF immunoaffinity column, which led the better purification effect and the lower limits of detection of the method ranging from1.0to8.0μg kg-1, the mean recoveries were between67%and107%. The developed method is suitable for the simultaneous detection of sulfonamides in food of animal origin.Chapter4:A method was developed for simultaneous determination of4kinds of β-agonists clenbuterol(CLE), salbutamol(SAL), ractopamine (RAC), and terbutaline(TER) in pork and beef by immunoaffinity chromatography coupled with ultrahigh performance liquid chromatography tandem mass spectrometry (IAC-UHPLC-MS/MS). The sensitivity and accuracy of the method were improved by optimizing the conditions of MS acquisition parameters and mobile phase, selecting the best injection solution and evaluating the matrix effect. The samples were cleaned by the IAC-AGON immunoaffinity column, which led the better purification effect and the lower limits of detection of the method ranging from0.2to3.5μg kg-1, the mean recoveries were between71%and96%. The developed method was applied to the detection of (3-agonists in actual commercial pork and beef.
