Banyan, Ficus microcarpa genera, is an important plant resource in Fujian. In order to develop and use this resource, extraction technology, Isolation and purified technology, antioxidant activity and acute toxicity effect of banyan leaves polyphenol were studied. Therefore, basic data for development and application of banyan leaves polyphenol in food processing were offered.(1) Effects of banyan leaves polyphenol extraction rate was influenced by five single factor experiments, they were ultrasonic power, ethanol concentration, extraction temperature, extraction time and solid-liquid ratio, which were studied in this paper. On the base of single factor experiments, the response surface methodology with three factors at three levels was designed according to the principles of Box-Benhnken experimental design of Design-Expert8.0.3software. The optional conditions of ultrasonic extraction were concluded as follows:ethanol concentration67.1%, ultrasonic power56.1W, ultrasonic temperature59.1℃extraction time40min and solid-liquid ratio1:20. The actual detection value of polyphenol extraction rate was15.3%, that is equal to predictable rate was96.2%.(2) Through the static adsorption and desorption experiments, NKA-9resin was selected as the material from6kinds of macroporous resins to purify the banyan leaves polyhenols. And dynamic properties of adsorption and desorption of the resin were studied. The results showed that the best adsorption conditions of banyan leaves polyphenols were as follows:the polyphenols concentration3.5mg/mL, pH4.0, ethanol concentration70%, flow rate3BV/h, elution rate5BV/h. In these conditions, the content of polyphenols in banyan leaves polyphnenols purified samples was75.3%.(3) Ultrasonic-assisted extraction of polyphenols from banyan leaves was studied in this paper, and NKA-9macroporous resin was selected to purify the banyan leaves polyhenols. The ability of scavenging hydroxyl radicals of banyan leaves polyphenols solution, inhibition of superoxide radical, getting rid of DPPH radicals, total antioxidant capacity, the activity of oil oxidation inhibition were studied. It turned out that the same mass concentration of banyan leaves polyphenols compared with Vc solution and tea polyphenols solution with stronger antioxidant capacity.(4) Acute toxicity effect of banyan leaves polyphenol was studied by Horn experiment. The results of acute toxicity experiments in Kunming mice of clean grade: there were not any poisoned symptom and death of mouse during14day-long observation periods. Banyan leaves polyphenol reached the actual non-toxic level according to the acute toxic dose graduation standard and blood routine indexes.
