BackgroundAbnormal lipid metabolism is one of the risky factors of atherosclerosis. Atherosclerosis is the main pathological basis of the cerebrovascular disease. Recent studies have shown that visfatin and leptin are related to the vulnerable plaque rupture, hypoxia, endothelial dysfunction, inflammation and lipid metabolism closely. Both of them can promote the occurrence and development of atherosclerosis.However, statin drugs cannot only make effects on the lipid-lowering, but also can play a role in anti-atherosclerotic through besides lipid-lowering effects such as anti-inflammatory and the improvement of endothelial cell function.Recent studies also have shown that statins may function as anti-inflammatory and improvement of endothelial cell by regulating the expression of visfatin and leptin.Therefore, the researches for visfatin and leptin in depth may provide new molecular targets for the study of atherosclerosis.ObjectiveThis study is aimed to give the high-fat and high-calorie diet for the production of rat atherosclerosis model.Through the observation of the activity of visfatin and leptin in visceral fat, it is aimed to find out the relationship between them.Combining investigating the expression of visfatin mRNA and leptin mRNA in atherosclerotic with exploring simvastatin prevention and treatment of atherosclerosis of the existence of possible mechanisms, this research is aimed at providing a theoretical basis for the clinical.MethodsRats were divided into four groups:normal control group,high-fat diet fed group, small dose simvastatin intervened group and large dose simvastatin intervened group. The normal control group rats were fed with common diet. The high-fat group and simvastatin intervened group were fed with high fat diet. After ten weeks, the rats’ abdominal aortas were sent to the pathological examination.Then the normal control group was fed common diet continuously, the high-fat diet fed group was lavaged the stomach with normal saline and the small and large dose simvastatin groups were lavaged the stomach with simvastatin respectively. After four weeks, RT-PCR was employed to study visfatin and leptin gene expression in viscera tissue of rats with different diets and effects of simvastatin on them.Results1.Compared with the normal control,the pathological section of the high-fat group shows atherosclerosis in it.2.Compared with the normal control, visfatin and leptin mRNA expression in the viscera fat tissue was increased significantly in high-fat diet group (P<0.05).3.The visfatin and leptin mRNA expression in the viscera fat tissue was decreased significantly in simvastatin intervened group compared with the high-fat group (P<0.05),and large dose simvastatin intervened group significantly decreased than the small dose group (P<0.05).Conclusion1.It is shown that both the high expression of visfatin and leptin in atherosclerotic rats’viscera adipose tissue can promote the occurrence of atherosclerosis development.2.The simvastatin can reduce the expression of visfatin and leptin in the atherosclerosis which caused by high-caloric and high-fat diet.It may be concluded that the simvastatin can be used to control the development of atherosclerosis.
