[Objective]1) To investigate distribution of HPV genetypes in local woman genital tract and to explore the relationships between multiple infections and different cervical lesions.2) To investigate the hr-HPV load and to analyse in uterine cervix with different pathological changes.3) To explore the relationships between integrated state of HPV16and different cervical lesions.[Methods]1) PCR-reverse dot blot was used to detect distribution of HPV genetypes in local woman genital tract.2) HC-Ⅱ was used to detect hr-HPV load in different cervical lesions.3) Multiplicitas real time was used to detect integrated state of HPV16in different cervical lesions.[Results]1) In shanghai area, the most common HPV types were HPV16(12.57%) and11(8.32%),followed by6(7.95%),52(6.56%),58(5.54%),18(4.71%),53(4.16%),33(3.60%), CP8304(3.14%),31(2.96%), and68(2.03%)2) Single and multiple infection rate was64.64%and35.36%, cervical cancer and high grade cervical precancerosis groups multiple infection rate was higher compared to other cervical lesions.(P<0.001)3) The loading dose of hr-HPV can discriminate the degree of cervical lesion, the higher the loading dose of hr-HPV is, the more impossible cervical cancer and high grade cervical precancerosis are. there was significant difference in different cervical lesions.(P<0.05)4) The rate of HPV16integration increased with the progression of cervical disease(0%、0%、22.22%、70%), the rate of HPV16dissociation depressed with the progression of cervical disease(90.72%、72.73%、33.33%、10%),the rate of mixed integrated HPV16in CINII+CINIII(44.44%) was significantly higher than that in other cervical lesions. In addition, the rate of integrated HPV16(pure integrated+mixed integrated) increased with the progression of cervical disease(9.28%、27.27%、66.67%、90%),the rate of HPV16integration was associated significantly with the degree of cervical lesion.(P<0.001)[Conclusion]In shanghai area,the most common hr-HPV types were HPV16、52、58、18, multiple infection was associated significantly with the cervical cancer and high grade cervical precancerosis. The loading dose of hr-HPV can discriminate the degree of cervical lesion, the higher the loading dose of hr-HPV is,the more impossible cervical cancer and high grade cervical precancerosis are.The rate of HPV16integration increased with the progression of cervical disease, the integration of HPV-DNA may be an early signal of cervical cancer. Detection of the condition of human Papillomavirus infection could be one method of evaluating cervical disease,the mothod can be usually utilized in clinical work,in addition,it can provide more reasonable project to the early diagnose of cervical cancer and early treat of cervical cancer.
