Establishment Of Rat Model With Helicobacter Pylori Associated Chronic Atrophic Gastritis

Objective:To develop a better rat model of Hp-associated chronic atrophic gastritis.Methods:Sixty grade two male Wistar rats were divided randomly into 4 groups,15 rats per Groups, i.e. model group (Hp, sodium salicylate and ethanol group), control group A (normal control group), control group B (Hp group), control group C (sodium salicylate and ethanol group), control group D(sodium deoxycholate, ethanol and ammonia solution group). The Hp-associated chronic atrophic gastritis models were induced with sodium salicylate, ethanol and culture broth of Hp(SSl)that were orally administered for 14 weeks. Both Hp bacteria cultivation, Gram staining of gastric antrum mucosal biopsy and rapid urease testing were made to detect the colonization of Hp, and to assess the histological indexes for gastric antrum mucosa.Results:Colonization of Hp in gastric mucosa was found in both model group and group B. The rats in model group, gastric antrum mucosa became thinner, neither glandular metaplasia nor atypical hyperplasia was found, but the numbers of glands in lamina propria decreased moderately, accompanying with moderate chronic active inflammation. The score (2.03±0.18,0.68±0.11,2.05±0.19) of chronic inflammation, active inflammation and decrease of intrinsic gland for model group were significantly higher than those in group A and group B (0.16±0.05,0.15±0.05,0; 0.39±0.11, 0.30±0.08,0;q=6.514-11.191,P<0.01), compared with group C and group D (1.95±0.19,0.54±0.08,2.02±0.19; 1.80±0.14,0.45±0.09,1.92±0.18), the difference was not significant (q=0.618-2.439,P>0.05)Conclusion:The features of rat model of Hp-associated chronic atrophic gastritis, made by the combined application of Hp with sodium salicylate and ethanol, are similar to those found in human subjects. Therefore, the model can be used as a better animal model for studying Hp pathogenicity and Hp-associated chronic gastritis treatment.
