The Expression And Clinical Significance Of P33^ING1Survivin、Id1in Gastric Cancer

Objective:To investigate the expression meaning and clinical significance of P33ING1, Survivin and Idl in human gastric cancer tissues and mutual relations among the three. At the same time, to analysis whether the expression of P33ING1, Survivin, Id1and the clinicopathologic parameters play a role in the prognosis of patients with gastric cancer.Methods:The protein expression of P33ING1was examined by western blot in20cases of gastric cancer tissues and corresponding adjacent tissues; at the same time, immunohistochemical staining method was used to detect the expression levels of P33ING1, Survivin and Idl in91cases of Paraffin sections of gastric cancer,30cases of normal gastric mucosa tissues,35cases of atypical hyperplasia tissues.Results:1) The positive rate of P33ING1was25.0%(5/20) and80.0%(16/20) in20cases of gastric cancer tissues and corresponding adjacent tissues by western blotting. There was significant statistical difference between them (P<0.05).2) Immunohistochemistry results showed that the positive rate of P33ING1, Survivin and Idl protein were41.8%(38/91),67.0%(61/91),63.7%(58/91) in91cases of gastric carcinoma. Compared to normal gastric tissues in30cases, atypical hyperplasia tissues in35cases, the differences were statistically significant (P<0.05). The expression of P33ING1, Survivin and Idl protein was significantly correlated with histological differentiation, lymph node metastasis, depth of tumor infiltration and clinical stages (P<0.05), but was not associated with patient’s age, sex, size of tumor, Lauren type and so on.3) The Spearman rank correlation analysis showed that there was negative correlation between P33ING1and Survivin (rs’=-0.208,P<0.05), also P33ING1and Idl (rs’=-0.-278,P<0.05). And there was positive correlationship between Survivin and Idl (rs’=0.281,P<0.05).4) The Log-rank test of survival curves and Cox proportional hazards regression model analysis showed that the depth of invasion and lymph node metastasis have relation with prognosis of gastric cancer, but the age, sex, tumor size, gross type, differentiation, Lauren classification and so on were not associated with prognosis of gastric cancer (P<0.05).Conclusion:1) The protein of P33ING1are abnormally lower expressed in gastric carcinoma. The protein of Survivin and Idl are abnormally higher expressed in gastric carcinoma. These may associated with occurrence, development, infiltration and metastasis of gastric cancer.2) The correlationship among P33ING1, Survivin and Idl suggested that they maybe play a synergistic action in the occurrence of gastric cancer.3) P33ING1, Survivin and Idl is expected to become the early diagnostic indicators and provide new therapeutic targets for the treatment of gastric cancer. They also may not be as the factors for assessing the prognosis of patients with gastric cancer.
