| Background: Stroke is very harmful to human health, has become oneof the important reasons for the human death, and often left differentdegrees of neurological dysfunction. Therefore, how to promote the neuralfunction recovery after stroke has become the key point of neurologicalrehabilitation research at present. The traditional Chinese acupuncturetreatment has been proved a positive therapeutic effect to recovery afterstroke, our initial studies have found that it has certain stimulative effect torepair damage function in cerebral ischemia area through electro-acupuncture(EA) of “Renzhong-Baihuiâ€(RB) or “Ganshu-Shenshuâ€(GS),the therapeutic effect was similar in both of the electro-acupuncture(EA)groups[1]. Whether the effect of combination treatment is better thanseparate treatment in promoting neural function recovery and theintervention of LINGO-1expression after stroke, it is worth furtherresearch and exploration.Objective: To compare the intervention effects of electro-acupuncture (EA) of RB with RB+GS on the neural function and the expression ofLINGO-1after focal cerebral infarction in rats.Methods: The healthy adult male SD (Spraguer-dawley) rats weresubjected to establish the model of middle cerebral artery occlusion(MCAO)by nylon monofilament suture, then were randomly divided intosham-operated group(n=8)ã€model group(n=32)ã€treatment group A(n=32)and treatment group B(n=32); the model group, treatment group A andtreatment group B were randomly divided into4sub-groups at1dã€3dã€7dand14d respectively after cerebral ischemia. The treatment group Aand thetreatment group B were respectively selected “Renzhong-Baihuiâ€(RB) and“Renzhong-Baihui+Ganshu-Shenshuâ€(RB+GS) as stimulating points, thetime of stimulation was30minutes once a day and all at10a.m.. The neuralfunctional recovery was evaluated by Longa,s score, RT-PCR andimmunohistochemistry were used to detect the expressions of LINGO-1mRNAand protein respectively in ischemic cortex.Results:â‘ The sham-operated group had no neurologicaldysfunction;â‘¡Neural function recovered significantly in the treatmentgroup A and the treatment group B compared with the modelgroup(P<0.05);â‘¢T he expressions ofLINGO-1mRNA and protein in themodel group reached the peak at1d and still higher than thesham-operated group at14d(P<0.01);â‘£The expressions ofLINGO-1mRNA and protein were lower in the treatment group A and the treatment group B than in the model group during7d14d(P<0.05); inaddition, the expression of LINGO-1protein in the treatment group B waslower than in the treatment group A, significant differences were foundbetween two treatment groups(P<0.05).Conclusions:â‘ T he expressions ofLINGO-1mRNA and protein inischemic cortex increased significantly;â‘¡EA may down-regulateexpressions of LINGO-1mRNA and protein after acute cerebral infarction,promote the neural functional recovery;â‘¢The therapeutic effect ofRB+GS group was better than RB group. |
