| Background:Psoriasis is a clinical common chronic recurrence skin diseases, psoriasis vulgaris is one of most commom types. At present,it is not completely that we know the cause of disease and pathogenesises of the psoriasis,the pathological features of psoriasis is the self-reactive T cells continued activates,so the immunity function abnormality is an important factor.Current common view is that under a certain genetic background, psoriasis is a kind of thymus derived cell mediated dermatitis,the abnormity of CD4+T lylmphocyte immunologic function is the most important. Psoriasis was generally considered to be a T1l/Th2cellular immune imbalance associated disease with a principal role for a long time. However,with the people to study in-depth found that only Th1/Th2cell imbalance theory can not fully explain the role of the immune pathogenesis of psoriasis.Therefore further reveal of the immune pathogenesis of psoriasis,improve and supplement Th1/Th2cell imbalance theory,to find effective treatment methods,need to study and explore new T cell subsets and cytokines,in order to better prevention and treatment of posoriasis.There are growing evidences that recently recognized two novel and unique subsets of CD4+T cells,T-helper17cells and CD4+CD25+regulatory T cells,also play important roles in the pathogenesis of psoriasis.Treg cells and Th17cells are new CD4+T cell subtype,which become a hot spot in the research years. They are original subsets distinguished from Thl and Th2cells. Treg cells are functioning in immunological regulation and immune suppression, Which play an important role in the maintenance of self immune tolerance. The alterations of Treg number and function are closely related to tumor,infection, autoimmune diseases and other diseases. FOXP3(Forkhead transcription factor p3) plays an important role in the formation of CD4+CD25+Tregs.It is the most specific mark of CD4+CD25+Tregs.Recent studies suggest that FOXP3may determine whether the treg cells have immunosuppressive function,that is it can reflect the level and functional activity of Treg to a certain extent;Th17cells,a new CD4+T cell subtype,have strong pro-inflammatory effects,highly convey IL-17,and excrete IL-6and TNF-a. Thl7has independent differentiate and growth regulatory mechanism which play an important role in immunological and infections disease.There is complex relationship between Treg and Thl7cells,the balance of Treg and Thl7cells plays important roles in tissue inflammation and autoimmune diseases.However, Thl7/Treg balance in PV has not been fully investigated and the conclusions is not yet uniform.In this study,we measured peripheral blood levels of Treg cells n FOXP3+Treg cells and Th17cells in psoriasis vulgaris patients, and the control group to observe the function of the Treg cells and Thl7cells in pathogenesis of the psoriasis vulgaris.lt can reveal the pathogenesis of the psoriasis and provide us another new way for the therapy of the psoriasis.Objective:To investigate the levels and significance of Treg cells and Th17cells in patients with psoriasis vulgaris (PV)Materials and Methods:1.34case diagnosed by clinic as psoriasis vulgaris patients who were selected in out-patient clinic in the second hospital of Shandong Medical University from June to October,2011. All cases are carried out period.Including18male patients,16female patients.The area and severity of skin was evaluated by PASI scoring; Patiens are exculded with other autoimmunity diseases and severe systemic diseases. All the cases involved had not taken any drugs that could affect function for three months such as corticosteroids and immuno suppressive agent,et al;18healthy donors were enrolled from health examination center as control, Including10male patients,8female patients. The control group do not have any family history and immunological diseases.With respect to the age,the gender,there was not significant difference between the two groups by analysis of t-test(p>0.05);2.Peripheral blood levels of Treg cellsã€FOXP3+Treg cells and Th17cells were measured by flow cytometry;3.By using SPSS17.0Statistical analysis software,Independent samples T-test was used to compare in two groups,P value of less than0.05was considered as significant;And pearson cor-relation was used to analysis correlation, P value of less than0.05was considered as significant. Results:1.The levels of Treg cellsã€FOXP3+Treg cells in the peripheral blood of Patients: There was a decrease in the percentage of Treg cells [(3.68±1.22)%vs (6.63±1.00)%,P<0.05] and FOXP3+Treg cells[(0.53±0.19)%vs (0.76±0.14)%P<0.05],the difference was statistically significant;2.The levels of Th17cells in the peripheral blood of Patients:There was a increase in the percentage of Th17cells[(2.20±0.78)%vs(0.65±0.22)%, P<0.05],the difference was statistically significant;3.Correlation analysis:The frequencies of Treg cells and FOXP3+Treg cells in patients with PV was positive correlation (r=0.563,P<0.05),but the frequencies of Treg cells in patients with PV was negatively correlated to frequencies of Th17cells (r=-0.522,P <0.05);but with no significant correlation to the PASI score and duration (with the score of PASI r=0.021ã€0.080,with the duration r=0.085ã€0.178,P>0.05)Conclusion:1.The levels of Treg cells in the peripheral blood of psoriasis vulgaris is lower,and the levels of transcription factor FOXP3+Treg cells is also lower,but The levels of Th17cells is higher;2.There was a significant negative correlation between Treg cells and Th17cells;3.The psoriasis vulgaris exist immune disequilibrium in Treg cells and Th17cells;The lack of Treg cells and The increase of Th17cells maybe participate the invasion of the psoriasis,this provides some scientific basis for investigating immune pathogenesis of psoriasis. |
