| Objective:To observe the effect of ethyl acetate extracted Chinese medicine sappan wood on the levels of cell factor IL-4in blood of rats with allogenic heart transplantion and the pathomorphology of heterotopic heart muscles, for the further study of sappan wood ethyl acetate extract on chronic rejection mechanism laying foundation.Methods:Choose the improved Ono mode and replicate the allogenic cardiac transplantation model. The allogenic cardiac transplantation model was established by using Wistar rats as donors and SD rats as receptors. After the success of the operation of continuous gavage for21days of35mg/(kg-d) CsA in rats, were divided into3groups randomly cyclosporine A group were given CsA according to12mg/(kg·d), sappan wood group were given sappan acetic ether extract according to736mg/(kg·d) and model group were given intragastric administration with olive oil depending on rats weight.After7days’dosing,the content of IL-4in blood serum was tested by ELISA,and heart myocardial pathological changes of heart transplant rats was observed.Results:Ethyl acetate extracted Chinese medicine sappan wood and CsA could obviously rise the levels of IL-4in blood of rats, the results are statistically significant (P<0.01). But Sappan wood group inferior to cyclosporine A group, the results are statistically significant (P<0.01) Compared with the model group, the sappan wood and CsA groups of rats showed obvious improvement in the pathologic changes in heart muscles, but Sappan wood group inferior to cyclosporlne A group.Conclusion:sappan acetic ether extract improves the pathologic changes in heart muscles, which gives rein to immunodepression probably by rising the secretion of IL-4. |
